Crabapple is widely used as Garden Courtyard Species in our country.Crabapple can not only burst into gorgeous flowers during spring,and when it comes into summer and autumn season,its fruits are mostly clustered and hung on branches,exquisitely carved,which is interesting and rich in ornamental value.Its fruit can be used not only for ornamental,but also has a large amount of active ingredients of polyphenolic flavonoids,which has significant antioxidant activity and great health care value for the human body.Fruits of three different varieties of crabapples?Malus micromalus,Malus‘Cinnabar'and Malus‘Royalty'?were used as experiment materials.Total phenolic contents?TPC?were determined using Folin-Ciocalteu reagent;total flavonoid contents?TFC?were determined using aluminum trichloride method;antioxidant activity?AOA?was assessed by DPPH and ABTS radical scavenging assays;individual phenolic compounds were determined by high performance liquid chromatography?HPLC?;antitumor effect?ATE?on human gastric BGC-803 cell lines was detected using Cell Counting Kit-8?CCK-8?and inverted microscope;in order to analyze the inhibition mechanism of BGC-803 cells,BGC-803 cells were detected by flow cytometry,and the relative expression of Bcl-2 and Bax,which are genes about apoptosis,was detected by RT-PCR.SPASS was used to correlate the total polyphenol content,antioxidant activity,and IC50 values of semi-inhibitory effects on gastric cancer cells BGC-803.The results showed that all the three extracts showed high TPC?TFC?AOA and ATE,and significant correlation was observed among TPC?TFC?AOA and ATE.;Malus‘Royalty'and Malus‘Cinnabar'are varieties with red flesh and showed higher AOA and ATE,and the deeper the color is,the stronger the activity is;The individual phenolics of five extracts mainly including procyanidins B1,procyanidins B2,?-?-epicatechin,chlorogenic acid and cyanidin galactoside,and high content of cyanidin galactoside and caffeic acid,as well as less content of cyanidin rutinoside were detected in Malus‘Royalty'and Malus‘Cinnabar',which were not found in Malus micromalus;The results of the detection of human gastric cancer cells BGC-803 by the polyphenol extract of Malus‘Cinnabar'fruit showed that with the increase of the extract concentration,the proportion of apoptotic cells increased significantly,and as the amount increased,the relative expression of the proapoptotic gene Bax was increased,while the relative expression of the apoptosis-inhibiting gene Bcl-2 was decreased.
