Crabapple,which belongs to the Rosaceae Malus,has been an ornamental plant for a long time.The studies of antioxidant activity about active ingredient of Crabapple are reported frequently,but there is no experimental research about antitumor activity,as well as the relativity analysis of active ingredient,oxidation resistance and antitumor activity.Four different fruit color Crabapples and Fuji apple as materials in this research.The species and contents of polyphenol and flavone,and the vitro antioxidant activity of extractive and the posion activity and strength in vitro on human esophageal cancer cells CaEs-17,the human gastric cancer cells BGC-803 and human colon cancer cells SW480 cells were analysised.The correlation of total polyphenols,flavonoids,antioxidant activity between antitumor activity were analysised.The results provide theoretical basis for comprehensive exploitation and utilization of crabapple in the future.The main research results are as follows:(1)Determined the types and content of mono-phenol of the test material by HPLC,the results showed that :the polyphenols in Fuji and Crabapples were about the same,but the content was different.Five kinds of materials methanol extraction were detected 12 kinds of mono-phenol,including Gallic acid,Procyanidins B1,B2,Epicatechin,Syringic acid,Phloridzin,Chlorogenic acid,Avicularin,Quercitrin,Rutin,Isoquercitrin.There were also discover Caffeic acid,p-Coumaric acid and Hyperin mono-phenol,content is low.But by comprehensive comparative analysis,the content of mono-phenol Crabapple is higher than the corresponding mono-phenol content in Fuji.(2)The methanol extraction of materials were examined in vitro antioxidant activity and analysized the correlation with the main free phenol,the results showed that: the total polyphenols and flavonoids,the DPPH and ABTS ability of scavenging free radicals of Crabapples were higher than Fuji.The red flesh crabapples(Malus ' Adams',Malus ' Red splendor')are higher the yellow flesh(Malus micromalus Makino).Malus ' Red splendor' has DPPH and ABTS strongest ability of scavenging free radicals,were 3831.91 mu mol,TE / 100 g,1763.23 mu mol,TE / 100 g.DPPH and ABTS ability of scavenging free radicals with four kinds of main mono-phenol(Gallic acid,Procyanidins,Catechin,Phloridzin)content are positively correlated relationship.The DPPH and Phloridzi content were extremely significant correlation was 0.964(p < 0.01);And ABTS with gallic acid,procyanidins,were extremely significant correlation relationship of,0.973 and 0.974,respectively(p < 0.01).(3)Five kinds of methanol extractions had different degrees inhibition of cell proliferation activity on SW480 cells,BGC – 803 cells and CaEs – 17 cells.The best concentration of five extractions of inhibition rate can reach about 70%.Analyize the differential significance of inhibiting effect on CaEs-17,SW480,BGC–803 by 5 alcohol extract under the same concentration,we can get the conclusion that the poison effect of tumor cells and intensity trend is the red flesh crabapples(Malus ' Adams',Malus ' Red splendor')> the yellow flesh(Malus micromalus)>Fuji;The cell morphological change has the positive correlation with the he poison effect of tumor cells and intensity:the poison effect and intensity are more stronger,the cell morphology changes more obviously.(4)The correlation about the polyphenols,flavonoids and antioxidant capacity and the antitumor activity of CaEs-17,SW480,BGC–803 were negatively related with IC50.The total polyphenols,flavonoids and antioxidant capacity had significant correlation with the IC50 of CaEs–17cells and SW480 cells.but the human gastric cancer BGC-803 cells showed a significant correlation,correlation coefficients were 0.926,0.937,0.933,0.909(p < 0.05).
