Huangqi Guizhi Wuwu Decoction On Diabetic Peripheral Neuropathy TRX And TXNIP Expression Effect

Objective:to study the huangqi guizhi wu wu decoction on diabetic peripheral neuropathy(DPN)model rats of therapeutic effect,and discuss the oxygen reduction sulfur protein oxidative stress system(Trx)and oxygen reduction sulfur protein interactions(Txnip)expression and the effect of the prevention and treatment of DPN new drug development and clinical application of Chinese traditional medicine to provide experimental basis.Methods:after screening qualified SPF SD rats,USES the abdominal cavity injection chain urea with cephalosporins 70 mg/kg(STZ)solution,preparation of DPN model.10 weeks after the screening model rats success only 45,randomly divided into model group(DPN),huangqi guizhi wuwu decoction group(group HQG)and a cobalt(JGA)amine group,each group of 15 only,in addition to select 15 as normal control group(NS group).Groups of rats lavage administration respectively,HQG group to the dose of 5 g/kg · d,JGA group to the dose of 170 mu g/kg · d,normal group and model group was given volume of distilled water.Groups of rats 1 times a day,for eight weeks in a row.Dosing in rats during the observation of general condition,body weight,water quantity and the change of the urine output,at the same time every 2 weeks once fasting glucose detection.After the dosing based,Western blot method is used to detect Txnip protein expression in rat sciatic nerve,USES the Real time PCR method to detect the rat sciatic nerve Trx mRNA and Txnip mRNA expression.Results:the general state of DPN model group rats is poorer,food intake,water quantity and the urine volume increased significantly,weight loss,"a little" sanduo symptoms,compared with NS group,p<0.05 or p<0.01;Compared with DPN group,HQG and JGA group general condition improved significantly,food intake,water quantity and significantly reduce the amount of urine,weight increased to different extent,p<0.05 or p<O.01.After compared with NS group,building groups of rats fasting glucose levels increased significantly,p<0.01;After medication,HQG and JGA group blood sugar levels have decreased,compared with DPN group,p<0.05 or p<0.01.Group,according to the results of Western blot and Real timePCR DPN rats sciatic nerve Txnip protein and gene expression significantly higher than that of NS group,HQG,JQA group was lower than that in group DPN P<0.01;DPN group rats sciatic nerve Trx gene expression was lower than that in group NS and group,JQA HQG group was obviously higher than that of the DP Ngroup,P<0.01).Conclusion:huangqi guizhi wuwu decoction can obviously improve DPN model rats general condition and symptoms,has a good therapeutic effect on it.Huangqi guizhi wuwu tang could inhibit DPN model in rat sciatic nerve Txnip protein and gene expression,to improve the level of Trx gene expression,thus reducing oxidative stress of peripheral nerve injury,to treat DPN.
