Study On Hepatoprotective Effect Of Hot Chinese Medicine Radix Aconiti Lateralis Preparata,Rhizoma Zingiberis,Pricklyash Peel

The drug property theory of Traditional Chinese Medicine(TCM)has been the centre theorical system Chinese medicine,a nd characteristics in Chinese Materia medica for thousands of y ears.Professor Kuang propose the new hypothesis which is base d on the scientific implication of“property and flavor theory of Traditional Chinese Medicine(TCM)":"Chinese medicine hast he property(gas)and flavor,the characteristics of characteristics of property(gas)and flavor.Flavor of a drug was related with s pecific efficacy.Property of a drug affects the body's energymet abolism and substance metabolism by different pass ways.Influe ncing therapeutic effects of drugs or lead to sideeffectassociated with biological effects."On the basis of complex analysis of the theory mentioned a bove,we study the property and flavor of Radix Aconiti Lateral is Preparata,Rhizoma Zingiberis,Pricklyash Peel Based on prot eomics.So we use the model of paracetamol induced acute liver inj ure in mice with the method intraperioneal injection was used t o study the protective effect of Radix Aconiti Lateralis Preparat a,Rhizoma Zingiberis,Pricklyash Peel.We find there have hep atoprotective effect.So,we use the model of paracetamol induc ed acute liver injure in mice with the method intraperioneal tos tudy the hepatoprotective effect of Radix Aconiti Lateralis Prepa rata,Rhizoma Zingiberis,Pricklyash Peel.We chose acetaminophen doses of 2.0g/kg,2.5g/kg,3.Og/kg,Result of lethal dose shows that acetaminophen doses of 2.5g/kg is the best doses of hepatic injury be made.After 7 days,fort y SD rats were randomly divided into five group and given by gavage once a day for 1week with Radix Aconiti Lateralis Prep arata,Rhizoma Zingiberis,Pricklyash Peel.After the last admini stration,each group were given acetaminophen doses of 2.5g/kg.Before the experiment,the rats were in abrosia 24 hours,but can ha ve water.Then we measure the level of ALB?ALP?ALT?AST?TBIL?TP,oxidative stress parameters in rats of GSH?MDA?SOD;histopathological featur;the protein expression of CYP1A2 and CYP2E1 of the liver in rats.Result shows that Radix Aconiti Lateralis Preparata can redu ced the level of ALP?ALT?AST?TBIL improved the level of ALB?TP;improved the level of GSH?SOD;reduced the level o f MDA,effectively reduced the hepatotoxicity;to determine the fl avon of CYP1A2?CYP2El,we find that the Radix Aconiti Latera lis Preparata can not decrease the flavon of CYP2E1 and CYP1A 2.Rhizoma Zingiberis can reduced the level of ALP?ALT?AST?TBIL improved the level of ALB?TP;improved the level of GS H?SOD;reduced the level of MDA,effectively reduced the hepat otoxicity;to determine the flavon of CYP1A2?CYP2E1,we find that the Rhizoma Zingiberis can not decrease the flavon of CYP 2E1 and C YP1A 2.Prickly ash Peel can reduced the level of ALP?ALT?AST?TBIL improved the level of ALB?TP;improved the level of GSH?SOD;reduced the level of MDA,effectively redu ced the hepatotoxicity;to determine the flavon of CYP1A2?CYP2E 1,we find that the Pricklyash Peel can not decrease the flavon o f CYP2E1 and CYP1A2.Acetaminophen as awide-used antipyretic analgesic drug,whe n in normal dose pill is safe without side effects.However,wh en taken in large doses,it can lead to serious liver toxicity.T here have been studies reported that many traditional Chinese m edicines have protective liver damage induced by acetaminophen.We found that:Radix Aconiti Lateralis Preparata,Rhizoma Zingi beris,Pricklyash Peel can decreased hepatic injury in rat serum ALP,ALT and AST activity and the level of TBIL;ALB,TP le vels be increased;also can effectively reduce oxidative stress r eaction and protect liver cells,reduce necrosis of the liver;but liver microsomesbody cytochrome P450 enzyme system in CYP1 A2 and CYP2E1 content had no significant effect,the specific mechanism needs to be further research.Summing up the above research results,we further confirme d the hot Radix Aconiti Lateralis Preparata,Rhizoma Zingiberis,Pricklyash Peel protective material foundation to the developmen t and clinical application of hot Radix Aconiti Lateralis Prepara ta,Rhizoma Zingiberis,Pricklyash Peel to provide theoretical b asis.
