Objective:1. To investigate the refrain role of proliferation on SGC-7901 cells acted by Matrine.2.To investigate the effects and mechanisms of Matrine combined with 5-FU for the growth of gastric cancer in nude mice implanted with gastric cancer cell SGC-7901.Methods:In vitro, Effects of Matrine on SGC-7901 cell proliferation was analyzed by MTS assay.In vivo,28 BALB/cA-nu mice, which had been inoculated human gastric adenocarcinoma cell line SGC-7901 subcutaneously,were divided into four groups at random and received treatments subcutaneously every day. The four groups were:control group(NS),Matrine group(Mat group),fluorouracil group(5-Fu group),Matrine+fluorouracil group(Mat+5-Fu group). We measured the volume of transplanted tumors every three days. After 28 days, the mice was killed and the tumors were immediately removed for further studies. We calculated the volume and weight of tumors. Imm-unohistochemistry was used to detect the expression of P53 and HIF-1α.Results:1.In certain range of concentration, matrine could obivously refrain Proliferationof SGC-7901 cells in a dose and time dependent manner.2.The xenograft volume and weight of experimental group was smaller than that of control group after the 4 weeks treatment,with statistical significance(P<0.05). 3. Immunohistochemistry showed the expression of P53 and HIF-la decreased(P<0.05).Conclusions:1.Martine could obviously inhibit the Proliefration of human gastric adenocarcinoma cell line SGC-7901.2.Martine and 5-Fu could inhibit the growth of cancer cell in vivo.3.Martine could decrease the expression of P53 and HIF-la protien.
