Our previous experiments obtained by traditional Chinese medicine Schisandra authentic Northeast,pesticide residues wind off the case study,organochlorine pesticides residues in schisandra contains hexachlorobenzene(HCB),?-BHC and p,p'-DDT,residual volume were 33.2ug/kg,63.1ug/kg,31.2ug/kg.Divaricate Saposhniovia Root proof herbs organochlorine pesticides residues containing ?-BHC,hexachlorobenzene(HCB)and p,p'-DDT,residual volume were 42.35ug/kg,42.35ug/kg,35.83ug/kg.According to Chinese Pharmacopoeia 2010 edition provisions Schisandra,Divaricate Saposhniovia Root herbs shall not exceed the total amount of BHC 200?g/kg,DDT total content shall not exceed 200?g/kg,HCB hexachlorobenzene total content shall not exceed 100?g/kg.From the measurement results of the sample can be seen:whether Schisandra pesticide residues BHC or off the wind,HCB and DDT alone are not exceeded national standards,but the wind and off Schisandra medicinal extracts obtained pesticide is sixty-six six,HCB and DDT mixture.In recent years,the majority of people dedicated to a single pesticide residues in toxicity studies,less toxic pesticides mixed research reports,and in the practical application of pesticides are mostly mixed after use of toxic pesticides on the human body after mixed harmful effects are additive or suppression is a question worth exploring.So this paper,windproof,HCH,HCB and DDT residues of pesticides containing Schisandra role in breast cancer cells,breast cancer cells to observe whether there is a case to be evaluated proliferation Schisandra,Divaricate Saposhniovia Root estrogenic effects of pesticide residues in the environment.The first experiment using MTT assay Schisandra,residual effects of pesticides on the wind off breast cancer cell growth;using Western Blot assay Divaricate Saposhniovia Root,Schisandra pesticide residues in breast cancer cells ERK1/2,p-ERK1/2 protein levels of expression,analysis of the mechanism of action of pesticide residues.MTT assay showed off the role of wind pesticide residues in breast cancer cells,administered 24 hours,the proliferation rate of 139.8%,compared with the control group,a significant difference(P<0.05).Administered 48 hours.the proliferation rate of 140.8%,compared with the control group,with a very significant difference(P<0.01).Administered 72 hours,the proliferation rate of 120.5%,compared with the control group,a significant difference(P<0.05);Schisandra Pesticide Residues role in breast cancer cells,administered 24 hours,the proliferation rate of 143.8%compared with the control group with a significant difference(P<0.05).Administered 48 hours,the proliferation rate of 144.5%,compared with the control group,with a very significant dif-ference(P<0.01).Administered 72 hours,the proliferation rate of 126.0%,there was a significant difference(P<0.01)compared with the control group.Western Blot results showed:Divaricate Saposhniovia Root proof pesticide residues in effect 48 hours after the breast cancer cells,cells,ERK1/2,p-ERK1/2 protein expression increased significantly different(P<0.05)compared with the negative control group,it was conclusions,Divaricate Saposhniovia Root pesticide Residues activation cascade in breast cancer cells through ERK1/2,when activated in breast cancer cells ERK1/2 phosphorylation,phosphorylated ERK1/2 is transported to the nucleus,acting on transcription factors in the nucleus,the regulation of target gene expression,and produce proliferation of breast cancer cells.Pesticide residues in Schisandra role in breast cancer cells after 48 hours,the cells ERK1/2,p-ERX1/2 protein expression increased,ERK1/2 and the negative control group with significant difference(P<0.05),p-ERK1/2 with the negative control group with significant difference(P<0.01).Therefore concluded that pesticide residues in Schisandra cascade activated by breast cancer cells ERK1/2,when activated in breast cancer cells ERK1/2 phosphorylation,phosphorylated ERK1/2 is transported to the nucleus,and transcription factor binding in the nucleus,regulates the expression of target genes,which produce proliferation of breast cancer cells,it causes reproductive toxicity.
