Objective Hepatocellular carcinoma(HCC)is one of the most common malignant tumors in the world.Its mortality rate is among the top five tumor-related causes of death.The liver,as the body's largest substantive organ,is also an important immune organ.In recent years,there have been many studies on the molecular immunity of the occurrence and development of hepatocellular carcinoma.The family of nucleotide binding domain receptor contains CARD domain-5(NLRC5)as a member of the NLRs family of pattern recognition receptors involved in the regulation of immune function,especially in the study of tumor immunity.To explore the NLRC5 in hepatocellular cacinoma(HCC)tissues,and to analysis the correlation with its expression,clinicopathological and prognosis of HCC.Methods The expression of NLRC5 was examined in 94 cases of HCC tissues and its adjacent normal peritumoral tissues by immunohistochemistry of SP.Westter blot was used to detect the expression of NLRC5 in 20 cases fresh HCC tissues and the corresponding normal peritumoral tissues.According to the expression of NLRC5,its relationship with clinicopathological features was analyzed.The 3-year overall survival rate of 94 patients was analyzed,and univariate and multivariate analysis were performed.Results The expression of NLRC5 was signficantly higher in HCC tissues(62.77%)compared with normal peritumoral tissues(19.15%,P<0.01).The positive expression of NLRC5 was related to the differentiation of hepatocellular carcinoma,TNM stage and cirrhosis.However,there was no correlation with tumor size,HBV infection,tumor capsule,sex and,age(P>0.05).The relative expression levels of NLRC5 protein was also significantly higher in HCC tissues(0.280±0.049)comparaed with corresponding normal peritumoral tissues(0.038±0.009,P<0.01).The 3-years overall survival of HCC patients with NLRC5 expression was significantly lower than that of patients without NLRC5 expression(P<0.01),and NLRC5 is an independent prognotic factor for HCC.Conclusion NLRC5 protein is highly expressed in hepatocellular carcinoma and is associated with the clinicopathological features,prognosis and other factors.NLRC5 is an independent risk factor for prognosis of hepatocellular carcinoma and may play an important role in the occurrence and development of hepatocellular carcinoma,It is expected to become a new tumor marker and potential therapeutic target for hepatocellular carcinoma.
