Effect Of Moxibustion Stimolation Of "Feishu"(bl13)and "Xinshu"(bl15)on Expression Of Myocardial Cell Autofhagy-assocoated Protein Bcl-2、bax、Fas And Fasl In Chronic Heart Failure Rats

Objective: To observe the possible mechanism of actions by moxibustion and explore the different interferences with general symptoms,ventricular mass index,cardiac function related index,concentration level of heart failure markers,and expression level of apoptosis related protein in myocardial tissue by two treatment groups,one of the groups selected the way of using moxibustion and captopril respectively while the other chose to combine moxibustion with captopril together.All mentioned above were carried out for the purpose of providing reliable experimental basis on the programme----the cure of CHF by moxibustion.Methods: Dividing Sixty male rats into twenty cages randomly and each includes 3 male rats,at the same time,selecting 4 cages as normal group while the rest were regarded as model.The model was established by intraperitoneal injection of ADR 2 mg/kg once a week for 12 weeks.The accomplished model rats were randomly divided into 4 groups that contained model group,moxibustion group,captopril group and the group combined moxibustion with captopril together;normal group and model group just fed these male rats without any steps;moxibustion group used moxibustion in rats of both Fei Shu points and Xin Shu points in the following 3 weeks,which was implemented 25 minutes per time everyday.According to the standard of 15 m L/kg,The captopril group was given a 5 mg/m L of captopril suspension,and the rats were treated with gavage per time everyday for three weeks.To record the rats’ changes of normal symptoms,heart rate,cardiac output,left ventricular systolic pressure and left ventricular end diastolic pressure,acquisition and major rat ventricular mass,enzyme-linked immunosorbent assay(ELISA)method to detect the rat serum brain natriuretic peptide(BNP)and N terminal pro brain natriuretic peptide(NT-pro the content of BNP,Bcl-2,Bax)level,Fas and Fasl protein expression were detected in myocardial tissue of rats with Western blot method after the treatment.Results: 1.The left ventricular and right ventricular mass index increased significantly in model rats(P<0.05),and heart function related indicators were significantly changed(P<0.01).After staining with hematoxylin and eosin(HE),it was found that the myocardial striations were unclear,most of the cells appeared swelling and vacuoles,and the changes of myocardial fibers were broken,dissolved and arranged in disorder.2.The behaviors of rats in each treatment group was improved,breathing and asthma,diet two stool,mental state and so on had different degrees of mitigation during the period of treatment.3.Ventricular mass index showed that the rats among moxibustion group,the captopril group,the group combined moxibustion with captopril together appeared decrease of their left and right ventricular mass index significantly(P<0.05).4.Statistics of cardiac function related indexes discovered that the rats’ cardiac function indexes of moxibustion group,captopril group and the group combined moxibustion with captopril changed significantly(P<0.05).5.The detection results of serum BNP,NT-pro BNP,BNP discovered that in model rats,the concentration of NT-pro BNP significantly increased(P<0.01);serum BNP concentration significantly decreased(P<0.01),moxibustion group and the group combined moxibustion with captopril serum NT-pro BNP concentration decreased(P<0.05),but serum NT-pro BNP concentration in captopril group decreased obviously.6.Western blot detection showed that the expression of apoptosis related protein Bax and Fas and Fas L all increased in model model group.The expression of Bcl-2 protein decreased evidently(P<0.01).The expression levels of Bax,Fas and Fas L reduced in all the treatment groups,and the expression of myocardial apoptosis protein Bcl-2 was up-regulated(P<0.01).Compared with moxibustion group and captopril group,the expression of combined group’s myocardial apoptosis related proteins Bcl-2,Fas,Bax and Fas L were different(P<0.01).Conclusion: 1.During the course of CHF treatment,moxibustion,captopril and moxibustion combined with captopril could inhibit the expression of apoptosis-related proteins Bax,Fas and Fas L in CHF rats,increase the expression of antiapoptosis protein Bcl-2,reduce myocardial injury,and have positive therapeutic effect on CHF.2.Moxibustion stimulation of "Fei Shu"(BL13)and "Xin Shu"(BL15)can significantly improve the symptoms of CHF rats and the heart failure,its mechanism may play similar to Captopril on apoptosis regulation,thus reducing myocardial injury.3.The combination of moxibustion and captopril in treating CHF rats is superior and more effective than single moxibustion or captopril in alleviating the symptoms,myocardial damage and heart failure.
