The efficacy of green plants with medicinal effects including traditional Chinese herbal medicine is generally the synergistic effect of various components,but its composition and content are vulnerably influenced by the ecological environment.Therefore,there are differences in the curative effect of the same medicinal materials from different sources.Fingerprint is a kind of effective analytical method based on the concept of overall identification of efficacy by macroscopically evaluating the quality and discriminating authenticity of traditional Chinese medicine.There are many chromatographic methods for establishing fingerprint,such as gas chromatography,liquid chromatography and thin layer chromatography.Compared to chromatographic methods,capillary electrophoresis technology has the characteristics of high separation efficiency,fast speed,simple operation,environmental protection and economy,making capillary electrophoresis stand out in many analytical methods.In this study,capillary electrophoresis was used to establish the fingerprints of Trifolium repens Linn,Momordica charantia L,Sonchus arvensis L.and plantaginis herba.The established CE fingerprints can provide new methods for quality evaluation of relative medicinal plants or traditional Chinese medicine.Objective:1.To establish capilllary electrophoresis fingerpint of Trifolium repens Linn with different origin.And all electrophorgrams of Trifolium repens Linn were analyzed by “Similarity Evaluation System for Chromatographic Fingerprint of Traditional Chinese Medicine Version 2004A”.2.The peak of chlorogenic acid was used as a reference peak to establish Sonchus arvensis L.capillary electrophoresis fingerprint,which provided a new method for the quality evaluation of Sonchus arvensis L.3.The capillary zone electrophoresis method was used to establish the fingerprint of Momordica charantia L.,which is both a kind of medicine and a vegetable.The overall quality of Momordica charantia L.in 14 producing areas was evaluated.4.The capillary electrophoresis fingerprint of plantaginis herba from different areas were established with the peak of quercetin-3-?-D-glucoside as reference peak.Similarity evaluation and clustering analysis were carried out to provide a new basis for the differences in plantaginis herba quality from different origins.Method:1.The conditions for the establishment of Trifolium repens Linn capillary electrophoresis fingerprint were: 50 mmol/L borax(pH 9.2)as background electrolyte and applied voltage of 12 kV,hydrostatic injection of 20 s(the hight of 11.5 cm),detection wavelength of 254 nm.2.The conditions for the establishment of Sonchus arvensis L.capillary electrophoresis fingerprint were: 20 mmol/L borax-0.5 mol/L boric acid-18 mmol/L?-cyclodextrin(pH 8.5)as background electrolyte and applied voltage of 15 kV,hydrostatic injection of 20 s(the hight of 11.5 cm),detection wavelength of 214 nm.3.The conditions for the establishment of Momordica charantia L.capillary electrophoresis fingerprint were: 30 mmol/L borax-45 mmol/L boric acid(pH 8.77)as background electrolyte and applied voltage of 15 kV,hydrostatic injection of 20 s(the hight of 11.5 cm),detection wavelength of 205 nm.4.The conditions for the establishment of plantaginis herba capillary electrophoresis fingerprint were: 30 mmol/L borax-40 mmol/L boric acid(pH 8.74)as background electrolyte and applied voltage of 15 kV,hydrostatic injection of 20 s(the hight of 11.5 cm),detection wavelength of 240 nm.Result:1.Capillary electrophoresis fingerprint of Trifolium repens Linn from differentareas was established.Ten common peaks were selected in the capillary electrophoresis fingerprint and one peak was identified.The similarity coefficient of Wenshang county and Jining city were 0.728,while other samples similarity coefficients were from 0.8 to 0.855.2.Capillary electrophoresis fingerprint of Sonchus arvensis L.from 11 different origins was established.Nine common peaks were selected as the fingerprint common peaks and three peaks were identified.The similarity coefficients of ten different habitats sample were over 0.9 except that of Pingyuan county,Dezhou city which was0.821.When the the genetic distance was 10,eleven different habitats sample were divided into two groups by cluster analysis.And all samples were divided into one categories when the genetic distance was 25.3.12 common peaks were selected in the Momordica charantia L.capillary electrophoresis fingerprint established by 14 areas Momordica charantia L.samples.The similarity coefficients of Momordica charantia L.capillary electrophoresis spectrogram from Yuncheng of Shanxi Province and Fuzhou of Fujian Province were from 0.80-0.85 while those from other 12 regions were greater than 0.85.The clustering results showed that all the samples were grouped into two categories when the genetic distance was 20.And all the samples were grouped into one categories when the genetic distance was 25.4.The capillary electrophoresis fingerprint of plantaginis herba was established with plantaginis herba sample from 12 areas and nine peaks were selected as commons peaks.The similarity evaluation and cluster analysis were used to analyse all samples.The similarity coefficients of plantaginis herba from 12 areas was from0.814 to 0.928.The clustering results showed that all the samples were grouped into two categories when the genetic distance was 10.And when the genetic distance was25,all the samples were grouped into one categories.Conclusion:The capillary electrophoresis fingerprints of several medicinal plants were established in this study.These methods are simple,accurate and reproducible.Chemical composition of samples from different regions enjoyed considerableconsistency,but contents of these constituents are affected by the ecological environment.
