Traditional Chinese medicine fingerprint (TCM-FP), as one of the quality control (QC) methods for TCM and its preparations, witnesses great development in recent years. Because the actual state of TCM-FP was based on the chemical compounds of extracts and their relative contents, so that there was a gap between the fingerprint and the curative effect which evaluate the quality of TCM truly. This study aimed at the actual state of TCM-QC, by combining the chemical compounds and the curative effect of TCM, developed a novel TCM-FP.This thesis mainly includes two parts.In the first part, the Chapter 2, the conception of"the functional TCM-FP"was put forward. Then the relative basic theories, the research methods were expatiated. And the advantages of Capillary Electrophoresis in the study of functional fingerprint of TCM were briefly introduced.The second part of this thesis consists of Chapter 3, 4 and 5, which is the main segment of the research. In this part, Ligustrum lucidum Ait., Gastrodia elata B1. and Rheum palmatum L were used as the research objects, and three different styles of functional TCM-FP were established.In the Chapter 3, following the principle and method on the measurement of oil-water partition coefficients by microemulsion electrokinetic chromatography (MEEKC), a novel TCM-FP was established. This method was applied to Ligustrum lucidum Ait., besides the establishment of its TCM-FP, the log Pow 3.63 of olesnolic acid which is one main active component in Ligustrum lucidum Ait. was obtained, too. And this TCM-FP was applied to guide the selection of solvents, for the extract of oleanolic acid from Ligustrum lucidum Ait.In the Chapter 4, took the different sources of Gastrodia elata B1. (Sichuan, Yunnan, Shanxi, Chongqing) as samples, the similarity of their TCM-FP was evaluated, before that, the running conditions of electrophoresis were optimized, and the methodology was also examined. At the same time, the relative content of gastrodin was examined, too. And, Gastrodia elata B1. distributed in Sichuan was used as an example, a functional TCM-FP was established, by which the log Powof compounds were measured. Further more, CE was used to explore the interaction between the extracts of Gastrodia elata B1. and BSA. In the Chapter 5, the complex reaction ofα-hydroxy anthraquinone and MgAc2 was utilized to distinguishα-hydroxy anthraquinone from the extracts of Rheum palmatum L, in order to explore a method to establish a novel TCM-FP, by which the function group may be screened.The study suggested that, the chemical compounds and curative effect of TCM were combined, through the functional TCM-FP. The identification and quality control of TCM were more complete and scientific.
