There are more than 7 million deaths from cancer worldwide every year,and this figure will rise rapidly.Non-small cell lung cancer(NSCLC)ranks first in global cancer deaths.Most patients are at advanced stages of diagnosis and have lost surgical treatment.Although clinical treatment has made great progress,the prognosis is still not satisfactory,and the 5-year survival rate is low.This article intends to explore the curative mechanism of two local non-small lung cancers in Yunnan Province in order to provide help for the diagnosis and treatment of lung cancer.In this paper,the effects of microRNA-218 on the proliferation,migration and apoptosis of human lung adenocarcinoma cells,lung squamous cell carcinoma cells and xuanwei lung adenocarcinoma cells were studied.(1)Detect the expression of miR-218 in several cell lines;(2)Use Real-time PCR and Western blot to detect the above three lung cancer cells transfected with miR-218 mimics.Expression levels of mRNA and protein of mid-miR-218 target genes PTEN,BCL-2,BMI-1 and YY-1;(3)Detection of miR-218 mimics after transfection with MTT proliferation assay and 3 cells scratch assay(4)The qualitative and quantitative analysis of the apoptosis of three lung cancer cells transfected by transmission electron microscopy and flow cytometry(FCM)were conducted.The results showed that the expression of miR-218 in NSCLC was lower than that of miR-218 in normal lung epithelial cells;after transfection of the mimics plasmid of miR-218,these three cells of lung cancer transfected with miR-218 The level increased,and the expression levels of the related tumor suppressor molecules PTEN and YY-1 increased,and the expression levels of the related oncogene BCL-2 and BMI-1 decreased.Compared with the blank control group,the function of each strain in the overexpression group was significantly decreased in growth,metastasis and the apoptosis was significantly increased.The results showed that the transfer of miRNA-218 mimics significantly inhibited the growth ability of NCI-H157,YTMLC-90 and XWLC-05,induced cell necrosis and apoptosis;transfection of miR-218 significantly inhibited the expression of BCL-2 and BMI1.Significant upregulation of PTEN and YY1 expression.It is speculated that the mechanism of miRNA-218 inhibiting lung cancer may be mediated through down-regulation of BCL-2 and BMI-1 genes and up-regulation of PTEN and YY1 genes.In addition,the analysis of the molecular mechanisms of lung cancer and these three lung cancers does not support the specificity of the lung squamous cell carcinoma(YTMLC-90)and Xuanwei lung adenocarcinoma(XWLC-05)from the perspective of molecular genetics.
