Recently,the ancient material silk from B.mori cocoons has been given a new luster in the biomedicine materials field for its advantage at mechanical property,biocompatibility,workability and degradability.Among these,preparation of functional silk fibroin nanoparticles and their application in drug delivery has been a popular topic.Nevertheless,the creation of other functional silk-based hybrid materials has been much less frequently.In this thesis,we firstly report a simple,versatile and efficient method for the encapsulation of different types of catalysts,including natural enzyme(glucose oxidase)and artificial enzyme(hemin)into biocompatible silk fibroin nanoparticles.Such semipermeable nanoparticles can act as nanoreactors,which facilitate complex reaction pathways under mild reaction conditions.Additionally,we test the possibility of the encapsulation of Saccharomyces cerevisiae(S.cerevisiae)cells in SF.It's expected to using this SF encapsulation to regulate the cells life cycles and cell stability.The main contents and results are as follows:1.Regenerated silk fibroin solution and various NPs,including pure SFNPs,SF-GOx NPs,SF-Hemin NPs and SF-GOx-Hemin NPs,were prepared and characterized FTIR spectra exhibited that the remarkable structure change of SF(from the random coil to the ?-sheet)was observed during the fabrication process of SFNPs.In addition,our results showed that the presence of these different catalysts had no significant effect on the morphology and structure of SF-based nanoparticles.2.When a catalytically-active species is encapsulated in these semipermeable SFNPs,they can be turned into a nanoreactor.The catalytical activities of SF-GOx NPs and SF-Hemin NPs were tested separately.3.Our results showed that SF-GOx-Hemin NPs had both glucose oxidase activity and peroxidase-like activity.Furthermore,this hybrid catalyst can catalyze a cascade reaction,include the oxidation of glucose to gluconic acid and H2O2 by oxygen,and the further oxidation of peroxidase substrates to color product by the H2O2.4.An exploratory work had been made to encapsulate S.cerevisiae cells through SF coating and microfluidics.The process conditions were optimized and the corresponding encapsulation result had revealed that the microfluidics-based method had more potential in the cells encapsulation with SF.
