Objective:According to differences of the main morphological structure in microimage of Saposhnikoviae Radix,Polygonatum odoratum and Asteris,Segmentation and quantitative characterization of different regions and features were realized with techniques such as image segmentation,image analysis,and the correlation between the main microscopic characteristics of medicinal materials and major components of the main pharmacodynamics was preliminarily analyzed,in order to establish quantitative method of a convenient and reliable on medicinal materials morphological identification and quality evaluation.And preliminarily to explore the histochemical localization of different medicinal materials,verify the color-reaction of the extracted components and establish the basis for the quantitative or semi-quantitative research of tissue chemistry.Methods:(1)Samples of a certain number of various medicinal materials were respectively an drandomly selected,Take the central sample from each,transverse microscopic slice were made by PEG 2000 as the embedding media and put it under microscope,then,each image was taken photo by using the the RuihogeRR300 C and ToupView software,while keep the overlaps of adjacent horizons,by using digital image processing technology and MATLAB software programming,three kinds of microscopic images of cross section of different medicinal materials were respectively stitching,Canny operator edge detection,binarization and segmentation of various microscopic characteristics and the analysis of its size,shape,number,distribution,and discriminant analysis of the data of the samples from the medicinal materials was carried out.SPSS 19.0 software was used to analyze relativity between the tissue morphologic feature of the medicinal materials and major componentsof the main pharmacodynamics measured by UV spectrophotometry.(2)Histochemical localization of three medicinal materials was carried out using colorimetric system consisting of 5% vanillin-glacial acetic acid and perchloric acid.Results:(1)The main microscopic characteristics in the transverse section images of the medicinal materials,such as phellem,phloem,xylem,oil-tubes and vessels,were respectively divided into sub-images;20 parameters were extracted from the sub-images,from which 14 parameters were screened out by stepwise discriminant analysis.Through discriminant analysis of the morphological parameters of Saposhnikoviae radix from different habitats,the total accuracy of the sample discrimination was 97.5%.In the correlation study,the relative content of the total chromogen ketone of Saposhnikoviae radix was respectively and negatively correlated with area fraction of the phellem and inner cavity of the oil-tubes.(2)The main microscopic characteristics in the transverse section images of the materials,such as epidermis,vascular bundle,mucous cell,were respectively divided into sub-images;37 parameters were extracted from the sub-images,from which 26 parameters were screened out by stepwise discriminant analysis.Through discriminant analysis of the parameters of Polygonatum odoratum from different habitats,the total accuracy of the sample discrimination was 93.8%.In the correlation study,the relative content of polysaccharides of polygonatum odoratum was positively correlated with area fraction of inner cavity of mucous cell.(3)The main microscopic characteristics in the transverse section images of the materials,such as epidermis and hypodermis,endodermis,vessels,were respectively divided into sub-images;16 parameters were extracted from the sub-images,from which 8 parameters were screened out by stepwise discriminant analysis.Through discriminant analysis of the parameters of Asteris from different habitats,the total accuracy of the sample discrimination was 71.2%.In the correlation study,the relative content of the total triterpenoids was not correlated with the morphological parameters of each tissue.(4)In histochemical coloration,the extractable component showed the corresponding color in the slice.Conclusions:This method can preferably segment and quantitatively describe microscopic characteristics of Saposhnikoviae Radix,Polygonatum odoratum and Asteris,and discriminate the fine difference of the tissue structure in the transverse section of the different samples,but also reflect the different origin of medicinal materials have specificity and stability of the species.Relative content of components of Chinese medicinal materials has a certain correlation with the characteristic parameters of its tissue structure,which is of certainreference significance for microscopic identification and quality evaluation of the medicinal materials from different habitats.
