ObjectiveThe effects of acupuncture and embedding "Baihui","Ganshu" and "Sanyinjiao" points on chronic unpredictable stress depression model(CUMS)were observed.Observe and analyze the changes of rat body weight,sucrose consumption and open field test in the process of modeling,and observe the changes of amino acid neurotransmitters in the hippocampus of experimental rats.Preliminary study on the principle of anti-depression by acupuncture and embedding,and the therapeutic effect of them.MethodsFifty male Sprague-Dawley rats,SPF grade,were randomly divided into blank group,model group,acupuncture group,catgut embedding group and fluoxetine group,10 in each group.One week after adaptive feeding,chronic unpredictable mild stimulation combined with solitary culture was used.The blank group rats were fed with 5 animals per cage without any stimulation.Rats in each model group were housed in single cages,and 7 days were used as a cycle.Each group received a series of stimulations for a total of 7 different stimulations for a total of 3 cycles.The total duration was 21 days.Model group does not receive any treatment after stress;Acupuncture group acupuncture "Baihui","Ganshu","Sanyinjiao" points;to bury the wire in the "Baihui","GanShu","Sanyinjiao" points of the embedding group;fluoxetine group is given oral fluoxetine.Each group of animals weighed on the first day,the 7th day,the 14 th day and the 21 st day of the enrollment,Day 1 and Day 21 record sugar consumption and open-air testing?All rats were killed after the experiment,the hippocampus was subjected to HE staining and the contents of GLU,ASP,GABA,and GLY were measured by sandwich enzyme immunoassay.ResultsBefore modeling,there was no significant difference in rats' weight between the each groups(P>0.05);as the experiment progressed,intervention measures began to play a role.Except for the model group,the rats' weight of each group showed a significant increase.The blank group increased significantly,while the weight gain of the model group was slow,which was significantly different from the blank group(P<0.05).Each group had statistically significant compared with the model group(P<0.05),and there was no significant different between with each group except the model group(P>0.05).The weight gain range of each group was blank group> fluoxetine group> acupuncture group> embedding group> model group.Before model establishment,there was no significant difference in sucrose consumption between the groups(P>0.05).After the experiment,except for the model group,there was no significant difference in sucrose consumption between the groups(P>0.05).The difference between the model group and the other groups was significant(P<0.05).There was no significant difference between the blank group,the acupuncture group,the embedding group,and the drug group(P>0.05).After 21 days of continuous modeling and treatment,the indexes of amino acid neurotransmitters in rats were detected by ELISA.The results showed that the concentrations of GLU and ASP in the model group were significantly higher than those in the blank control group(P <0.05).The concentrations of GLU and ASP in the acupuncture group,embedding group and fluoxetine group were significantly higher than those in the control group(P <0.05).Compared with the blank group,the concentrations of GLU and ASP in the acupuncture group,embedding group and fluoxetine group were both lower than those in the blank group,and there was no significant difference between the two groups(P> 0.05).Compared with the model group,the concentrations of GABA and GLY in the other groups were all significantly increased(P <0.05).Compared with the blank group,the concentrations of GABA and GLY in the model group were significantly decreased Statistical significance(P <0.05),acupuncture group,embedding group and fluoxetine group three treatment groups no significant difference(P> 0.05).Conclusion1.Acupuncture and embedding has the effect of improving behavioral metrics such as body weight,sucrose water consumption,vertical movement,and horizontal exercise activity in depressed rats.There is no significant difference between the acupuncture group and the embedding group,but it is significantly different from the model group.It shows that the effect of acupuncture and embedding is comparable,both of which can alleviate the physical and physiological dysfunction of depression.2.Acupuncture and embedding can reduce the concentration of GLU and ASP in depressed rats,increase the concentration of GABA and GLY,and effectively improve depression.3.The antidepressant mechanisms of acupuncture and embedding may be related to amino acid neurotransmitters.
