Projective:In order to investigate the mechanism of the Strengthening-Brain-and-Tranquilizing - Mind Acupuncture to the post stroke depression(PSD),we have observed the effect on the body weight,behavior and contents of monoamine neurotransmitter in hypothalamu for the rat with post stroke depression(PSD),before and after Strengthening-Brain-and- Tranquilizing -Mind Acupuncture.Methods:1.Divide the rats randomly into 6 groups as:blank array,model one,acupuncture one, fluoxetine one and acupuncture-fluoxetine one.2.Produce the animal model with disease-verification,i.e.the rat model with Post stroke depression(PSD) by combining the cerebral ischemia caused by removing blood vessel from the cerebral cortex with chronic unpredictable mild stress(CUMS) and separation.3.Measure the body weight,content of sugar water drunk,and score of the open-field test under the chronic unpredictable mild stress(CUMS) for each rat on the 1st day,the 8th,the 15th,the 22nd,and the 29th,observe the effect of the Strengthening-Brain-and-Tranquilizing -Mind Acupuncture on the body weight,consumption of the sugar water,and the score of open-field test.4.Execute the rats in full on the thirty sixth day.Observe the contents of monoamine neurotransmitter(dihydroxyphenyl ethylamine,5-hydroxytryptamine,noradrenaline)in hypothalamus of the PSD rats model with enzyme linked immunosorbent assay(Elisa).Results:1.Observe the effect of the Strengthening-Brain-and-Tranquilizing-Mind Acupuncture on the body weight and the consumption of the sugar water of the rat with the post stroke depression(PSD):In comparing with the blank array,the body weight and the consumption of the sugar water of the model array was decreased;In comparing with the model array,the body weight of the acupuncture array,acupuncture-fluoxetine one and fluoxetine was increased,the consumption of the sugar water was increased,And there is no difference between the acupuncture array and fluoxetine one.and the score of the open field test was higher. 2.Observe the effect of the Strengthening-Brain-and-Tranquilizing-Mind Acupuncture on behavior of the rat with the post stroke depression(PSD):The score of the movement levelly and vertically are all less than the blank array's.The acupuncture array can increase the score of the movement levelly of the PSD rat quickly,but the effect on the fluoxetine array was the best one;The acupuncture array can increase the score of the movement vertically of the PSD rat,and the effect was better than the others.3.The effect on the contents of 5-hydroxytryptamine,dihydroxyphenyl ethylamine,noradrenaline in hypothalamus of post stroke depression rat model after Strengthening-Brain-and-Tranquilizing-Mind Acupuncture.'s treatment:The content of dihydroxyphenyl ethylamine have no impact.The content of 5-hydroxytryptamine is much higher(P<0.01),but the fluoxetine array is better.The content of noradrenaline is much higher(P<0.01),but the fluoxetine and acupuncture array is the best.Conclusion:1.By using the complex modeling method,combining the cerebral ischemia caused by removing blood vessel from the cerebral cortex with chronic unpredictable mild stress (CUMS) and the separation,the animal model with the post stroke depression(PSD) can be produced successively.2.The result of the detection by this institute indicates that:by improving the contents of monoamine neurotransmitter in hypothalamus,the Strengthening-Brain-and-Tranquilizing -Mind Acupuncture is able to improve the symptoms of the rat with the post stroke depression (PSD).3.The Strengthening-Brain-and-Tranquilizing-Mind Acupuncture is an effective method to treat the post stroke depression(PSD) and it is worth to be researched and developed further.
