ObjectiveTo explore the effect of acupuncture on the behavior and the degree of neurological deficit in the model rats of post stroke depression(PSD),to explore the effect of acupuncture on the content of neurotransmitter and nitric oxide synthetase in the cerebral cortex of the rats with ischemic PSD,to evaluate the effect of acupuncture on the improvement of limb motor disorder and depressive symptoms caused by hypoxic-ischemic brain injury;to observe the effect of acupuncture The effect of acupuncture on the brain gut pathway of rats with ischemic PSD was studied by the theory of"no neuron injury",and the effect of acupuncture on the brain gut pathway of rats with ischemic PSD was clarified The mechanism of cognitive affective disorder in rats.MethodsShort-term middle cerebral artery occlusion(MCAO)was used to establish a right cerebral artery embolization reperfusion model(temporary middle cerebral artery occlusion/reperfusion,tMCAO/R),lasting for four days after surgery,a post stroke depression(PSD)model was established with a combination of chronic loneliness and chronic unpredictable mild stress(CUMS),only two stimuli per day(10 in total,two stimuli per day per random-numbered chart),for 21 consecutive days.Give PSD rats acupuncture intervention(GB13 and GV24)and drugs(paroxetine hydrochloride tablets,according to the dose of 10 mg/kg lavage dosing)intervention,intervention once a day,intervention in 14 days in a row,in the process of intervention consistently give CUMS stimulation.All the rats were observed respectively in MCAO made die before,CUMS model,7 acupuncture intervention and acupuncture intervention in 14 days after detection.The experiment was divided into three parts:(1)to evaluate the effect of acupuncture on the behavior(open field experiment,sucrose preference rate)and the degree of neurological deficit in rats with ischemic PSD;(2)to observe the effect of acupuncture on the content of neurotransmitters(5-HT,Da,NE)in cerebral cortex,serum and colon tissues of rats with ischemic PSD by using ELISA detection technology;(3)to observe the effect of acupuncture on the content of neurotransmitters(5-HT,DA,NE)by using ELISA technology To observe the effect of acupuncture on the content of eNOS and iNOS in cortex,colon and serum of rats with ischemic PSD,to evaluate the effect of acupuncture on the level of oxidative stress in brain gut pathway of rats with ischemic PSD,and to elucidate the mechanism of acupuncture regulating brain gut pathway to inhibit neuron damage and improve cognitive and emotional disorders of rats with ischemic PSD.Results(1)According to the results of open field experiment and sucrose preference experiment,compared with the normal group,the level activity times,vertical activity times and sucrose preference rate of rats in the ischemic PSD model group were significan tly reduced(P<0.001),suggesting that the depression behaviors and symptoms were obvious after the model was established,while the level activity times,vertical activity times and sucrose preference rate of rats in the acupuncture group were significantly lower than those in the model The rats in group A were significantly higher than those in group B(P<0.001).The symptoms of decreased activity and interest in paroxetine group were also relieved.Compared with the normal group,there was no significant difference in the score of neurological deficit in the sham operation group(P>0.05).The score of neurological deficit in the ischemic PSD model group was significantly higher(P<0.001),suggesting that the sham operation could not replace the MCAO ischemia-reperfusion operation,which caused neurological deficit in the ischemic PSD model Compared with the PSD model group,the nerve function of the sham acupuncture group had no significant change(P>0.05),and the nerve function of the sham acupuncture group had no significant difference(P>0.05)The difference was statistically significant(P<0.001).The difference was statistically significant(P<0.05).It indicated that acupuncture could effectively improve the neurological deficit of rats with ischemic PSD,but paroxetine was not satisfied with the neurological deficit of rats with ischemic PSD.Compared with the model group,the scores of motor function,depressive symptoms and neurological deficit in the 3-mA group were significantly worse.Compared with the 3-mA group,the degree of neurological deficit and depressive symptoms in the acupuncture+3-mA group were significantly reduced,suggesting that acupuncture may have a regulatory effect on the oxidative stress level of neuron brain gut pathway in the ischemic PSD rats,and it may be through reducing neuronal oxidative second part of the experimental results showed that compared with the sham operated rats,the contents of 5-HT,Da,NE in the cerebral cortex of the rats in the ischemic PSD group were significantly reduced(P<0.01,P<0.001,P<0.01),the contents of 5-HT,Da,NE in the colon were significantly reduced,and the differences were statistically significant(P<0.01,P<O.001,P<0.01),the contents of 5-HT,Da,NE in the serum were decreased,The difference was statistically significant(P<0.05,P<0.05,P<0.05),which indicated that the content of neurotransmitters in cerebral cortex,colon and serum decreased significantly after hypoxic-ischemic brain injury,which may be the cause of cognitive emotion abnormality and depressive behavior in rats with ischemic PSD.Compared with rats in PSD model group,the neurotransmitters in brain cortex,colon and serum in sham acupuncture group were significantly decreased The contents of 5-HT,DA and NE in the cortex of acupuncture group increased significantly(P<0.001,P<0.001,P<0.001),the contents of 5-HT,DA and NE in the colon increased significantly(P<0.001,P<0.001,P<0.001),the contents of 5-HT,DA and NE in the serum increased significantly(P<0.001,P<0.001,P<0.001)The significance(P<0.01,P<0.01,P<0.01)showed that acupuncture can effectively improve the content of neurotransmitters in cerebral cortex,colon and serum of rats with ischemic PSD,and it is beneficial to the recovery of HPA axis stability,the improvement of cognitive and emotional ability of rats,and paroxetine can also improve the depressive symptoms of rats with ischemic PSD,and the mechanism may be related to promoting the increase of neurotransmitter content.The content of neurotransmitters in 3-mA group is less than that in 3-mA group,while the content of neurotransmitters in acupuncture+3-mA group is increased,which suggests that inhibition of autophagy flow can cause abnormal secretion of neurotransmitters,and the increase of neurotransmitters may be due to the promotion of autophagy flow,so as to improve the neural function defect and cognitive and emotional abnormality caused by hypoxic-ischemic brain injury.(3)In the third part of the experimental study,we found that compared with the sham operated group,the content of eNOS and iNOS in the cerebral cortex of the rats with ischemic PSD increased significantly(P<0.01,P<0.001,P<0.01),the content of eNOS and iNOS in the colon increased significantly(P<0.01,P<0.001,P<0.01),and the content of eNOS and iNOS in the serum increased significantly(P<0.01,P<0.001,P<0.01)The results showed that the oxidative response was enhanced and the neurons were damaged,which led to the loss of nerve function.At the same time,the cognitive emotion was abnormal and the depression symptoms after stroke occurred.The contents of eNOS and iNOS in the cortex of acupuncture group were significantly reduced(P<0.001,P<0.001,P<0.001),The differences were statistically significant(P<0.001,P<0.001)The difference was statistically significant(P<0.001,P<0.001,P<0.001).The contents of eNOS and iNOS in serum were significantly lower(P<0.01,P<0.01,P<0.01).It indicated that acupuncture could effectively improve the content of neurotransmitters in cerebral cortex,colon and blood serum of rats with ischemic PSD,which was beneficial to the recovery of HPA axis stability and the improvement of cognitive and emotional ability of rats Good,paroxetine can effectively improve the depressive symptoms of ischemic PSD rats,and the mechanism may be related to the inhibition of oxidative response,the promotion of neurotransmitter secretion and the restoration of HPA axis balance.The content of eNOS and iNOS in cerebral cortex,colon and serum of 3-mA group increased,while that of eNOS and iNOS in acupuncture+3-mA group decreased,which indicated that acupuncture might save autophagy,restore autophagy flow and inhibit oxidation reaction,so as to protect the secondary injury of neurons,promote the repair of nerve function of brain defect and improve depression symptoms.ConclusionThe damage of neurons caused by oxidative reaction and the abnormal secretion of neurotransmitters caused by HPA axis disorder are the key factors to cause cognitive and emotional disorders and emotional disorders in rats with ischemic PSD.Acupuncture can effectively improve the depressive symptoms of rats with ischemic PSD.The basic reason is that acupuncture can promote the increase of neurotransmitter content in rats with ischemic PSD,at the same time,inhibit the oxidation reaction to protect neurons from secondary damage.This process may be related to the activation of autophagy flow of neurons.We also found that the change trend of neurotransmitters and NOS in cerebral cortex and colon was the same.We speculated that the brain gut pathway might exist in the process of neural function defect and cognitive emotion abnormality in rats with ischemic PSD.It might be involved in the mechanism of acupuncture inhibiting oxidation reaction,promoting the increase of neurotransmitters and improving depressive symptoms of ischemic PSD the role and role of the approach in it need to be further explored in the future.
