Phellodendron amurense is a kind of perennial woody plant which combines medicine,economy and environment.It is an indispensable medicine for the processing of Chinese medicinal herbs.It has multiple functions such as antibacterial,heat and dry dampness,catharsis,detoxification,and jaundice.Some studies have shown that the extract of berberis has antioxidant effect,but the antioxidant effect of different extracts in different parts has not been reported in detail.With the melioration of economic conditions and the improvement of the quality of life,health care has been paid more attention,and the kinds of antioxidants have emerged.Synthetic antioxidants are cost-effective,but they are also reported to be damaging to health,and natural antioxidants are gaining popularity.In order to further develop and utilize the antioxidant function of berberberry resources,the antioxidant effect of berberis seeds,leaves and bark was compared in this experiment.This thesis select amur cork tree as the research object,in order to radical scavenging ability as the size of the antioxidant capacity evaluation standard in vitro,screening the amur cork tree seeds,leaves,bark coarse separation antioxidants,hoping to phellodendron amurense antioxidant activity research results applied to the natural antioxidant,delay aging,disease prevention and control of drug research and development,etc.The main results of this study are as follows:1,USES the ultrasonic assisted extraction technology,using ethanol solution and pure water leaching of phellodendron amurense,extraction times,ethanol concentration,leaching solution leaching liquid ratio,extraction temperature,ultrasonic time and so on the influence of conditions of phellodendron amurense antioxidant active substances extraction,investigation,using orthogonal test with four factors three levels for the amur cork tree to determine the optimal extraction process,it is concluded that the best extraction technology conditions as follows:extraction number two,70%ethanol concentration,extraction liquid ratio in 25 g/mL,extraction temperature 40?,ultrasonic time is 2 h.2,The amur cork tree seeds,leaves,bark of crude extract,water extract and alcohol extract were determined,the results showed that water extract and alcohol extract has certain antioxidant capacity,and the alcohol extract of antioxidant capacity than water extract.3,According to the principle of different polarity of different organic solvents,using organic solvents in turn petroleum ether,ethyl acetate,n-butanol crude extract of phellodendron amurense liquid-liquid extraction separation,and petroleum ether extraction phase respectively,ethyl acetate,n-butanol extraction phase extraction phase and water phase.Experiment in vitro antioxidant(removal of DPPH free radical,hydroxyl free radical,FRAP method,the ability of removing ABTS free radicals)the ability of analysis,the results show that the extraction layers have certain antioxidant capacity,comprehensive comparison:ethyl acetate phase removal ability is stronger than n-butanol phase,and stronger than the water phase,therefore petroleum ether phase ability of scavenging free radicals is weakest.
