BackgroundTea is a traditional Chinese drinks,the habits of drinking tea and tea culture have been handed down.There are many kinds of tea.Some are the compatibility ingredients of traditional Chinese medicine treatment,other is the diet treatment of traditional Chinese medicine.It is still controversial that the effect of tea in gastric mucosa injury and protection.Most studies have found that the tea and its extract has improve the defense ability of gastric mucosa,antitumor,anti-oxidation,hypoglycemic lipid-lowering,protecting brain,preventing oral disease,etc.However,some people who don't often drink tea or drinking tea while limosis would have abdominal discomfort,nausea after drinking tea.There is no evidence that gastric mucosa has directly damaged by tea.How to illustrate the relationship between tea and the mechanism of gastric mucosa defense is still needing the system method in animal experiments accepted by the international.Protection mechanism of the stomach can be divided into two categories:mechanical barrier and chemical barrier.The detection index also can be divided into two categories:anatomical and biochemical index.In anatomy,most of the study is still using the theory of mechanism of gastric mucosa protection published by H?jgaard L[7]in 1991.It said that the protection mechanism of the gastric mucosa could be divided into three layers:gastric mucus barrier,gastric mucosa barrier,gastric mucosal blood flow.According to this theory,the research at home and abroad considered the thickness of gastric mucus and the gastric mucosa,gastric mucosal blood flow as evaluation of the anatomical index in gastric mucosa.In addition,the composition of loose bed and the fixed layer in gastric mucous layer is also commonly used as anatomical index.In biochemical indicators,it would be measured by prostaglandin E?PGE?and the content of amino sugar in the stomach.Prostaglandin?prostaglandins?plays an important role to the protection of gastrointestinal system,which is recognized as the protection factor of endogenous gastric mucosa.The effect and function of mucosal barrier is closely related to the content of amino hexose.Determination of the amino hexose in gastric mucosa is considered as the evaluation of function of gastric mucous membrane barrier,the investigation of pathogenesis in peptic ulcer disease,the important biochemical index of determination of drugs effect.Moreover,monitoring serum pepsinogen of diagnosis and treatment in gastric disease can reflect the state of gastric mucosa,forecasting the occurrence of atrophic gastritis and gastric cancer,which can also be used to determine the efficacy of helicobacter pylori eradication and reflect the secretion of gastric acid.In addition,the recent study found that all kinds of damage or stress factors caused stomach movement dysfunction is one of the important factors of gastric mucosa damage.Multiple experiments showed that gastric contraction continued to a period of time,which could appear erosion or ulcer injuries of creases in the gastric mucosa.Also,some scholars research showed that the gastric contraction movement disorder caused gastric emptying disorder and duodenal gastric reflux,which is an important factor of gastric ulcer occurrence.Therefore,this research through the determination of the thickness of the gastric mucous,prostaglandin E2,amino hexose,pepsinogen I and pepsinogen II and gastric emptying rate as the evaluation index of the impact of different concentration of green tea in gastric mucosa.In conclusion,this study based on the above background,through giving different concentrations of green tea extract for 4 weeks in the Sprague Dawley?SD?rats.We observe the thickness of gastric mucus after disposed to green tea extract,prostaglandin E2,amino hexose,pepsinogen I and pepsinogen II level and gastric emptying rate,etc.The research investigates if different concentration of green tea extract would protect the stomach or harmful effects of the body,which would provide a healthy diet advice for the people.ObjectiveBy feeding the green tea extract of male SD rats for 4 weeks,we investigated whether different concentrations of green tea extract have different effects on gastric mucosal protective barrier and gastric emptying in rats,and which concentration of green tea protects the role of gastric mucosa,which concentration is adversely affected,as well as the impact of various concentrations on gastric emptying,so as to provide the general population with healthy dietary recommendations,in order to guide people the correct way of life and prevention methods.MethodsA randomized controlled study was used.A total of 48 male SD rats?Sprague Dawley?weighing 150-180 g were used in this experiment.Six rats in each group were randomly divided into 1.5 g group,3 g group,6 g group and blank control group.Those groups were fed with 1.5g,3g,6g tea and 200ml tap water respectively.When feeding 4weeks,the gastric mucus thickness,gastric acid secretion,PGE2,serum pepsinogen,hexosamine,and gastric emptying rate were measured by appropriate reagents and instruments after fasted for 48 hours.After the experiment was completed,the data was input into the Excel table of the computer in time,and statistical analysis was performed by SPSS 17.0 statistical software.Measurements are expressed as meansąstandard errors of the mean.Data were analyzed using a one-way analysis of variance?ANOVA?,with any differences between means assessed by LSD-t test to evaluate statistical significance.Statistical significance was indicated by P values of<0.05.Results?A?Anatomy indicators:?1?The measurement of gastric mucus thickness:The gastric mucus thickness in the blank control group was the largest,the tea group at different concentrations was slightly lower than the blank control group,the difference was statistically significant in the 3g group and the 6g group compared with the blank control group(P3g=0.025,P6g=0.02,all<0.05).However,there was no significant difference between the 1.5g group and the blank control group(P1.5g=0.131>0.05),and no significant difference was in the experimental groups.?2?The degree of gastric mucosal inflammation:There was no significant inflammatory reaction in the gastric mucosa of the blank control group.With the increase of tea concentration,the degree of inflammatory reaction in gastric mucosa slightly increased.The mean score of 1.5g group was 2.5,3.0g group was 4.8,6.0g group was 6.0,inflammation score of each group was higher than the blank control group,1.5g group,3.0g group,6.0g group was significantly difference compared with the blank control group?P<0.05?.The difference was statistically significant in 3.0g group,6.0g group when compared with 1.5g group?P<0.05?.However,there was no significant difference between 3.0g group and 6.0g group?P>0.05?.?B?Determination of biochemical indicators:?1?Determination of gastric acid:The total amount of gastric acid in the blank control group was the least,and the total amount of gastric acid also increased slightly with the increase of green tea concentration.However,there was no significant difference between the two groups when compared with each other?P>0.05?.?2?Prostaglandin E2?PGE2?:The concentrations of PGE2 between groups was not significantly different?P>0.05?.?3?Determination of serum pepsinogen:The concentrations of PG I and PG II between groups were not significantly different?P>0.05?.?4?Determination of hexosamine:The content of hexosamine in the gastric mucosa of the blank control group was the highest,while different concentration of green tea groups was slightly lower than the blank control group.There was significant difference in both the 3g group and the 6g group compared with the blank control group(P3g=0.012,P6g=0.005,all<0.05),but the difference between 1.5g group and blank control group was not statistically significant(P1.5g=0.056>0.05),and there was no statistical difference among the experimental groups.?C?Determination of gastric emptying rate:The gastric emptying rate of the blank control group was the lowest,and the gastric emptying rate of the tea leaves in different concentrations groups were higher than that of the blank control group.There was significant difference in the 1.5 g group,the 3 g group and the 6 g group compared with the blank control group?P<0.001,P<0.05,P<0.05?,and the rate of gastric emptying increased with the increase of green tea concentration.The differences between 3 g group and 1.5 g group were statistically significant?P=0.007?.The differences between 6g group and 1.5g group were also statistically significant?P<0.001?.However,when the concentration of tea was further increased,there was no statistical difference between 6g group and 3g group?P=0.098>0.05?.ConclusionIncreasing the concentration of green tea can reduce the thickness of gastric mucous layer in rats,aggravate the inflammation of gastric mucosa,can reduce the content of hexose in rat gastric mucosa.The increased concentration of green tea has a certain damage on gastric mucosal defense barrier.Increasing the concentration of green tea can increase gastric emptying rate.The results are correlated with concentration.
