Cervical cancer is the most common malignant tumor in the female reproductive system,The younger incidence of cervical cancer poses a great threat to young and middle-aged women..Application in our country medicine anticancer plants has a long history,from the plant medicine anti-cancer active ingredients is an important part of anticancer agents.Among them,baicalin and wogonin are representative components of the plant drug Scuthllaria baicalensis,which have a good inhibitory effect on cervical cancer.However,the anti-cervical cancer effect and mechanism of the compound combined with baicalin and wogonin have not been studied in depth.In this paper,the effect and mechanism of the combination of baicalein and wogonin on the apoptosis of cervical cancer Hela cells were investigated.In this study,Hela cells were cultured in vitro and treated with drugs.The negative control group and the compound group were set,and the drug concentration points were50,100,150,200,250,300 and 350 ?g/ml.MTT assay was used to explore the inhibitory effect of the complex on cell proliferation,and the relationship between baicalein and wogonin on Hela cells and the optimal concentration of the combination were obtained.Hoechst 33258 method was used to observe the apoptosis morphology after the compound intervention.Annexin V-FITC/PI was used to quantitatively detect the apoptosis rate after the compound intervention.Western Blot was used to detect the relative expression of apoptosis-related proteins in endoplasmic reticulum pathway and mitochondrial pathway.In this paper,we studied the relationship between baicalein and wogonin in combination,explored the role of complex in inducing apoptosis,and analyzed the mechanism of baicalein-wogonin complex(1:1)inducing Hela cells apoptosis.This study showed that the compatibility of baicalein and wogonin 1:1 had a synergistic effect.The optimal concentration of baicalein combined with wogonin was200 ?g/ml,which could increase the apoptotic rate of Hela cells.Its mechanism may be the interaction between Bcl-2 family and Caspase family,which induces Hela cells apoptosis through mitochondrial pathway and endoplasmic reticulum pathway.This studyprovides an experimental basis for the study of baicalein-wogonin complex in inducing apoptosis of Hela cells in cervical cancer.
