ObjectiveThis study aimed to detect the expression levels of interleukin-9,interleukin-6 and tumor necrosis factor-? in colonic mucosa of patients with ulcerative colitis by immunohistochemistry,and analyze its relationship between clinicopathological factors,so as to investigate the roles of IL-9,IL-6 and TNF-a in the occurrence and development of ulcerative colitis,and provide reference for clinical diagnosis and treatment.MethodsRetrospective study method was used in this research.Selected from December 2013 to December 2015 Binzhou People's Hospital outpatient and hospitalized with complete clinical and pathological data of ulcerative colitis patients in 60 cases.They were divided into mild group(n=38)and severe group(n=22)according to their disease conditions,whereas 20 healthy people served as control group.The expression of IL-9,IL-6 and TNF-a were observed under light microscope,and the results were determined by immunohistochemical staining.And to analyze the relationship between sex,age and severity of clinical and pathological factors.Results1?The cytoplasm and(or)cell membrane of IL-9,IL-6 and TNF-? positive cells was brownish yellow or light yellow in color,and the positive cells are mainly lymphocytes.2?The levels of IL-9,IL-6 and TNF-a in mucosa of each group were compared:the expression levels of IL-9,IL-6 and TNF-a in colonic mucosa of UC patients were all significantly higher than those in control group,and the differences of which were statistically significant(P<0.01).3?The levels of IL-9,IL-6 and TNF-a in mucosa of patients with ulcerative colitis in different severity were compared:the percentage of IL-9,IL-6 and TNF-a positive cells in moderate-severe UC group was all higher than that in mild UC group,and the differences of which were statistically significant(P=0.0 15,0.019,0.038,respectively).4.Analysis of clinical and pathological factors:Age:The percentage of IL-9,IL,6 and TNF-a positive cells in colonic mucosa of patients with ulcerative colitis in 20-49 years old group were significantly higher than those in the groups of under 20 years old and over 49 years old,the differences of which were statistically significant(P values are:0.001,0.002,0.028).Sex:There was no statistical significance in the percentage of IL-9,IL-6 and TNF-a positive cells of UC groups between female and male(P values are:0.936,0.878,0.713).Conclusion1?The levels of IL-9,IL-6 and TNF-? have been highly expressed in colonic mucosa of patients with ulcerative colitis,and the expression levels are all increased with the severity of the disease.2?The expression of IL-9,IL-6 and TNF-a in colonic mucosa of patients with ulcerative colitis was correlated with age,but its expression level was not related to gender.3?IL-9,IL-6 and TNF-a can be indicators to monitor UC disease severity and efficacy,which contribute to the clinical diagnosis and treatment options of UC,perhaps to provide new treatment for UC opportunistic,especially to the supply a reliable basis for the diagnosis of patients who have not yet suffered from obvious clinical symptoms.
