Objective:To investigate the relationship between the single nucleotide polymorphism?SNP?rs1050244 C>T of the 3'untranslated region?3'UTR?of damage specific DNA binding protein 2?DDB2?gene and the genetic susceptibility of hepatocellular carcinoma?HCC?in Guangxi population,to explore the pathogenesis of HCC.Methods:A hospital-based case-control study was conducted in 1073 patients with HCC and 1119 cancer-free matched controls from January 2007 to April 2011 in First Affiliated Hospital of Guangxi Medical University and Affiliated Tumor Hospital of Guangxi Medical University.Sequenom Mass ARRAY technique was used to genotype the 3'-UTR region of DDB2 gene SNP rs1050244 C>T.?2 analysis was used to compare the general data of the study subjects between the case group and the control group.Multivariate logistic regression was used to calculate the Odds ratios?OR?and 95%Confidence intervals?95%CI?of the SNP genotypes and HCC risk and their interaction with the environment.Results:The results showed that no significant statistical differences in the distributions of age grouping,sex was found between cases and controls;However,smokers,drinkers,and hepatitis B virus?HBV?infection individuals in cases group are significantly higher than the controls?P<0.05?.After adjusted by age,sex,smoking,drinking status and HBV infection,no statistically significant correlations in respective SNP and the risk of HCC were found.Under the dominant genetic model,stratification analysis was further carried out according to factors such as age grouping,gender,ethnicity,smoking,drinking and HBV infection,It was found that compared with those carrying rs1050244CC genotype in uninfected HBV population,The risk of HCC was significantly lowerinCT/TTgenotypes?OR=0.31,95%CI=0.13-0.72,P=0.006?,Gene-environment interaction analysis showed that there was an interaction between the uninfected HBV population and the CT/TT genotype(P interaction=0.002).Conclusions:The rs1050244 polymorphism in the 3'-UTR region of the DDB2 gene is significantly associated with HCC susceptibility,but this association is only present in non-HBV infected individuals.The results of this study can be further validated in larger sample populations.
