ObjectiveIn this study,the effects of gemcitabine combined with radiotherapy on the proliferation inhibition,apoptosis and migration of Tca8113 cells were studied by in vitro study of human tongue squamous cell carcinoma Tca8113 cells,and the effects of gemcitabine on the radiosensitivity of human tongue squamous cell carcinoma Tca8113 cells and their possible correlations were investigated.Mechanism,providing a theoretical basis for the treatment of tongue cancer.MethodsTongue squamous cell carcinoma Tca8113 cells were purchased from the Shanghai Institute of Biotechnology Cell Bank.Human tongue squamous cell carcinoma Tca8113 cells were cultured in vitro,and human tongue squamous cell carcinoma Tca8113 cells were treated with different concentrations of gemcitabine(0,0.001,0.01,0.02,0.03,0.1,1?mol/L)for 24 h,and the cell proliferation activity was determined and the minimum onset concentration was selected.After that,different irradiation doses(0,2,4,6 Gy)were combined with gemcitabine to treat Tca8113 cells for 24 h.The cell survival rate was determined by MTT method,and the subsequent experimental irradiation dose was determined.The follow-up experiments were divided into four groups: blank control group,gemcitabine group,radiotherapy alone group and gemcitabine combined with radiotherapy group.Plate colony formation assay was used to study the colony forming ability of each group.Hoechst33342 staining method was used to observe the morphological changes of Tca8113 cells in each group.The effect of gemcitabine on apoptosis of Tca8113 cells induced by radiotherapy was observed by flow cytometry.Western blot was used to detect gemcitabine combined with radiotherapy.The effect of gemcitabine on the migration of Tca8113 cells by radiotherapy was examined by the effect of gemcitabine on the expression of bcl-2,bax,Cleaved caspase-3 and Cleaved caspase-9 in Tca8113 cells.ResultsThe minimum inhibitory effect of gemcitabine on proliferation inhibition of Tca8113 cells was 0.02?mol/L.The viability decreased with increasing concentration(P<0.05).The minimum effective radiation dose is 4Gy.The colony formation experiment showed that the cell colony forming ability of the gemcitabine combined with radiotherapy group was weakened and was in the stationary phase compared with the radiotherapy alone group.Hoechst33342 staining showed that the cell size was reduced in the gemcitabine group,the radiotherapy group,and the gemcitabine combined with the radiotherapy group.Bright blue,cell apoptosis,apoptosis in gemcitabine combined with radiotherapy group;flow cytometry further confirmed that the apoptosis rate of gemcitabine combined with radiotherapy group was significantly higher than that of radiotherapy group,and increased the radiosensitivity of Tca8113 cells;Western Compared with the radiotherapy alone group,the expression of bax protein in gemcitabine combined with radiotherapy was negatively correlated with the expression of bcl-2,and the expression of Cleaved caspase-3 and Cleaved caspase-9 was more significant.Gemcitabine combined with radiotherapy significantly inhibited the migration of Tca8113 cells,and the migration ability was weaker than that of the radiotherapy alone group.ConclusionsGemcitabine can reduce the cell viability of tongue squamous cell carcinoma and inhibit the proliferation of Tca8113 cells in a concentration-dependent manner with a minimum effective concentration of 0.02 ?mol/L.Low concentration of gemcitabine can inhibit cell proliferation and promote apoptosis in Tca8113 cells after radiotherapy,and inhibit the migration of Tca8113 cells.Therefore,gemcitabine combined with radiotherapy can effectively improve radiosensitivity,and the two may have synergistic anti-tumor effects.Provide relevant data for further basic experimental research and lay the foundation for clinical application.
