ObjectiveTo investigate the expression of TRPM1 in the human tongue squamous cell carcinoma tissue and cell line Tca8113 .Methods(1) Immunohistochemical staining was used to survey the expression of TRPM1 in the human tongue squamous carcinoma tissue and the normal tongue tissue.(2) Immunocytochemistry was used to survey the expression of TRPM1 in Tca8113 cell line.(3) Western-blot was used to survey the expression of TRPM1 in the human tongue squamous carcinoma tissue Tca8113 cell line and the normal tongue tissue.(4) Reverse transcription polymerase chain reaction(RT-PCR) was used to survey the expression of TRPM1 mRNA in Tca8113 cell line and the normal tongue tissue.(5) The data were analyzed with SPSS 11.0 software package.Results(1) The result of Immunohistochemistry indicated a high expression of TRPM1 in 23 of 25 cases of the human tongue squamous carcinoma tissue, but 12 of 15 cases in the human normal tongue tissue were no expressed. The rates of positive expression in the two tissues were 92% and 20%. So the expressions of TRPM1 between the two tissues had significant difference ( 0.05).(2) The enhanced expression of TRPM1 was observed in the Tca8113 cell line from the result of Immunocytochemistry.(3) The expression of TRPM1 in the human tongue squamous carcinoma tissue and Tca8113 cell line is higher than that of the normal tissue by Western-blot.(4) RT-PCR showed higher expression of TRPM1 in Tca8113 cell line than that of human normal tongue tissue.ConclusionThere may be a close relationship between the expression of TRPM1 and the process of the tongue squamous cell carcinoma...
