ObjectiveTo investigate the expression of LRG-1 in carcinoma of tongue and cell line Tca8113,and the influence of LRG-1 on proliferation of HUVEC and angiogenesis in vitro,analyze the relationship between LRG-1 expression and clinical pathological parameters,to provide a theoretical basis for further research LRG-1 and its mechanism in the pathogenesis of tongue cancer.MethodsThe expression of LRG-1 in 40 cases of tongue squamous cell carcinoma(TSCC)and normal tissue beside TSCC,20 cases of tongue atypical hyperplasia,and 20 cases of tongue cancer in situ,were detected by S-P Immunohistochemical method;The expression of LRG-1 in cell line Tca8113 were examined by flow cytometry;The expression of LRG-1 in the human TSCC tissue and cell line Tca8113 were detected by western blotting.The influence of LRG-1 on proliferation of HUVEC was determined by MTT assay;The effects of LRG-1 on angiogenesis were detected by Matrigel tube formation assays.SPSS 17.0 package was fitted to this study.Results1.LRG-1 expression is mainly localized in the cytoplasm,and is also seen in nucleus.2.The positive expression of LRG-1 in human TSCC tissue(85%,34/40)was significantly higher than the normal tissue beside TSCC(10%,4/40),the invasive tongue cancer(30%,6/20),and the tongue cancer in situ(50%,10/20)(P<0.05).3.The expression of LRG-1 in human TSCC tissue was not associated with the patients' age and sex,but correlated with the clinical stage,degree of differentiation and lymph node metastasis(P<0.05).4.The expression of LRG-1 in cell line Tca8113 is also high.5.LRG-1 can promote HUVEC proliferation and angiogenesis in vitro.ConclusionsThe expression of LRG-1 in the human TSCC tissue and cell line Tca8113 is high,which was not associated with the patients' age and sex,but correlated with the degree of differentiation,clinical stage and lymph node metastasis.LRG-1 can promote HUVEC proliferation and angiogenesis in vitro,which suggests that it may play an important role in tumorigenesis and development through promoting tumor angiogenesis.
