| Objective: To investigate whether TGF-?1 mediates the anti-inflammatory effect of acupuncture on adjuvant arthritis rats.Methods: Eighteen rats were randomly divided into complete Freund's adjuvant model group(AIA),needle module(AIA+MA),needle model + TGF-?1 receptor blocker group(AIA+MA+T?R bloker),each Group 6 only.The model of AIA was an intradermal injection of adjuvant on the right side of the rat.The intervention method was acupuncture at the bilateral Zusanli.In the AIA+MA+T?R bloker group,the right foot of the rat was injected once every other day after modeling,and the PBS was injected into the AIA group and the AIA+MA group with the anterior plantar.During the experiment and after taking the material,the effect of the foot volume was measured by the foot drainage method.After the samples were taken,the expression levels of TGF-?,CD86,MCP-1,IL-1? and 2iNOS in the paws of rats were detected by real-time fluorescent quantitative PCR.Results:(1)The results of foot drainage test showed that the paw swelling of AIA+MA rats remained at a relatively low level during the experiment.The paw swelling of AIA rats was higher than that of AIA+MA rats during the whole observation period.At the two time points of d9 and d13,the swelling of AIA rats was significantly higher than that of AIA+MA rats.The difference was statistically significant.Significance(P<0.05).The paw swelling of AIA+MA+T?R bloker rats was also higher than that of AIA+MA rats during the whole observation period,and the difference between the two groups was statistically significant(P<0.01).The difference between the two time points of d13 was statistically significant(P<0.05).(2)The results of RT-qPCR showed that the expression of MCP-1 mRNA in AIA+MA+T?R bloker rats was significantly higher than that in AIA+MA,the difference was statistically significant(P<0.05),and AIA was higher than AIA+.The trend of MA.The expression of TGF-? mRNA in AIA and AIA+MA+T?R bloker was lower than that in AIA+MA.The expression of IL-1? and 2iNOS mRNA in AIA and AIA+MA+T?R bloker was higher than that in AIA+MA.However,there was no statistical difference(P>0.05).There was no significant difference in the expression levels of CD86 mRNA between the three groups.Conclusion:(1)TGF-?1 receptor blocker can block the effect of needle to some extent,indicating that TGF-?1 is involved in the process of acupuncture treatment of adjuvant arthritis.(2)TGF-?1 may mediate the anti-inflammatory effect of acupuncture by down-regulating MCP-1. |
