Objective:To study the chemical constituents of the petroleum ether and EtOAc extracts from the Illicium verum Hook.F.and their inhibitory effects on tumour necrosis factor-a(TNF-a)production in lipopolysaccharide(LPS)-stimulated mouse macrophages(RAW 264.7)cells.Methods:The petroleum ether and EtOAc extracts of the fruits of Illicium verum Hook.F.were isolated and purified by using normal-phase and reversed-phase silica gel chromatography,middle pressure liquid chromatography and high performance liquid chromatography.The structures of the isolated compounds were identified by NMR spectroscopy.The isolated compounds were assayed for their inhibitory effects on TNF-a production in LPS-stimulated RAW 264.7 cells by ELISA method.Results:Eighteen compounds were isolated from the petroleum ether and EtOAc extracts of I.verum.The structures of compounds 1-18 were identified as anisic acid(1),1,4-benzenediol(2),?-hydroxybenzoic acid(3),2,4-dihydroxybenzoic acid(4),1'-(1-methoxyphenyl)-1',2',3'-trihydroxypropane(5),1'-(1-methoxyphenyl)-l'-propanol(6),1-ethoxy-4-methylbenzene(7),1-methoxy benzaldehyde(8),(1'S,2'S)-l'-(l-methoxyphenyl)-propane-1',2'-diol(9),(1'R,2'S)-1'-(1-methoxyphenyl)-propane-1',2'-diol(10),verimol A(11),verimol B(12),harmandianone(13),1R,3R,4R-trihydroxy-menthane(14),?-sitosterol(15),bis(2-propyloctyl)phthalate(16),bis(2-propylheptyl)phthalate(17)and bis(1-methylpentyl)ester(18).Among them,compounds 2,6,7,9,13,14,15,16-18 were isolated from the family Magnoliaceae for the first time.Compounds 11 and 13 could significantly inhibit the secretion of TNF-a in LPS-stimulated RAW 264.7 cells. |
