Study On Chemical Constituents From The Resin Of Populus Euphratica

Resina Populi is a natural resin secreted from Populus euphratica of the Salicaceae family.P.euphratica is a precious forest resource in the desert,which has the characteristics of heat resistance,drought resistance and saline-alkali resistance and is distributed in western Inner Mongolia,Gansu,Xinjiang and other places in China.The resins from P.euphratica is a small number of medications and Uyghur medical drugs.The earliest recorded in"Tang Bencao”describes its characteristics as stomach salt,bitter cold,non-toxic,into the stomach,mainly appling for the treatment swelling and pain in throat,tuberculosedadenitis,duodenal ulcer and other diseases.Our research group has been engaged in the researc:h of traditional Chinese medicinal resin for a long time.The literature found that there is little study on the chemical composition of Resina Populi,and the material basis for its efficacy is not clear.It shows that the chemical composition of Resina Populi has yet to be further explored and the material basis of the efficacy of the drug needs to be clarified.By extracting with 95%EtOH,with the use of silica gel,MCI gel CHP 20P,Sephadex LH-20,RP-18,HPLC various chromatographic separation methods,and combining with multiple spectroscopy techniques,we have isolated and identified 56 compounds,including 19 new ingredients.The isolated compounds mainly include abietane-type diterpenes,cembranoid-type diterpenes,verticillane-type diterpenes,and a few triterpenoids.A part of the compounds were subjected to activity screening,and the results showed that compound 1 and compound 18 were cytotoxic.Thus,compounds 1 and 18 were examined for their cytotoxic activities against human cancer cells including Kyse30,HepG2,A549,BGC-823,and MDA-MB231,respectively.And compared with HUVEC normal human umbilical vein endothelial cells.The results showed that compounds 1 and 18 display low cytotoxicity against normal cells and appreciable cytotoxicity in cancer cells,with 18 to be more sensitive in HepG2 cells with an IC50 value of 27.0μM.This topic is the first time to study the chemical constituents of Resina Populi,which provides scientific basis and reference for further research and elucidation of the material basis of traditional efficacy of Resina Populi.
