| The theory of medicinal properties of traditional Chinese medicine is one of the core of the basic theory of traditional Chinese medicine.In a narrow sense,the medicinal properties of traditional Chinese medicine refer to the four properties of cold,heat,temperature and coolness.Although the ideas and methods in the study of the medicinal properties of modern traditional Chinese medicine can distinguish the four properties of traditional Chinese medicine to a certain extent.However,most of the studies on the use of modern science to explore the theory of the four characteristics of traditional Chinese medicine are divorced from the overall concept of the basic theory of traditional Chinese medicine.The ultra-weak luminescence of life is the embodiment of the metabolic level of the body and has integrity.In the early stage of our laboratory,we explored the quantification of the medicinal properties of traditional Chinese medicine based on the ultra-weak luminescence of Scenedesmus.In order to further study,on the basis of the previous research,the cold and hot properties of traditional Chinese medicine were determined based on the research method of ultra-weak luminescence detection of Scenedesmus.The spontaneous luminous intensity of traditional Chinese medicine was measured,and an experimental platform for studying the cold and hot properties of traditional Chinese medicine was established based on the ultra-weak light emission detection of mice,and the quantification of the medicinal properties of traditional Chinese medicine was discussed from different angles.The advantages and disadvantages of the two experimental methods were compared to explore the feasibility of quantification of traditional Chinese medicine.Objectives:Based on the determination of the cold and hot properties of traditional Chinese medicine based on the ultra-weak luminous detection of Scenedesmus,the accuracy and repeatability of this method were discussed,the ultra-weak luminous intensity of traditional Chinese medicine mice was detected,and the parameters which could characterize the cold and hot properties of traditional Chinese medicine were explored.An experimental platform for studying the cold and hot properties of traditional Chinese medicine based on ultra-weak light emission detection in mice was established,and the correlation between the parameters of ultra-weak light emission detection in mice and the biochemical indexes of liver was analyzed to verify the experimental results of ultra-weak light emission detection in mice.This paper tries to find out the reason why some of the cold and hot properties of traditional Chinese medicine have not been identified successfully based on the ultra-weak photoluminescence detection of Scenedesmus.To compare the advantages and disadvantages of the method of studying the medicinal properties of traditional Chinese medicine based on the ultra-weak luminescence detection of Scenedesmus spp and based on the ultra-weak luminescence of mice,and to explore the objective quantitative index of the medicinal properties of traditional Chinese medicine.Contents:1.Determination of cold and hot properties of traditional Chinese medicine based on ultra-weak luminescence of Scenedesmus.Angelica sinensis,aconite,turmeric,perilla stem,fairy grass,Scutellaria baicalensis,honeysuckle vine,Gardenia jasminoides,Artemisia annua,Fructus Artemisia,Gaoliang ginger,Cistanche deserticola,Weiling were selected from 220 traditional Chinese medicines determined by Scenedesmus biologica biological indicator method in our laboratory.Immortals,longan meat,Corydalis,Rhizoma Corydalis,Chinese rose,maple skin,calendula,Millennium Health,Anemarrhena,Tianmendong,Radix Isatidis,plantain,semen Lepidii,Leucaena,moutong,Platycladus orientalis leaves,Gentiana,The stability and repeatability of the cold and hot properties of traditional Chinese medicine based on the ultra-weak luminescence of Scenedesmus sp.,as well as the reliability of the previous determination results,were discussed by repeated determination of these 30 traditional Chinese medicines.2.A preliminary study on the detection of cold and hot properties of traditional Chinese medicine based on ultra-weak luminescence in mice.First of all,the relevant experimental conditions were explored,including the week age,body weight,coat,detection site,intragastric concentration,dose and time,anesthetic dose,dark adaptation time,instrument detection parameters,data processing and analysis,and so on.Then,based on the results of repeated determination of the medicinal properties of 30 traditional Chinese medicines based on the ultra-weak luminescence of Scenedesmus,the mice were given intragastric administration and their spontaneous luminescence was detected to study whether there were rules and differences in the bioluminescence indexes of mice after the decoction of different medicinal herbs were perfused into the stomach.The results were compared with the results of repeated determination of ultra-weak luminescence of Scenedesmus.3.Determination of liver related biochemical indexes in mice by intragastric administration.Biochemical indexes are related to the metabolic level of the body,and the ultra-weak luminescence of life is also related to the metabolism of the body,so the relevant biochemical indexes of mouse liver are used to verify the experimental credibility of ultra-weak light emission detection of traditional Chinese medicine in mice.The mice were killed by intragastric administration of ultra-weak light emission detection,and the liver was taken out.The activities of superoxide dismutase(SOD),total antioxidant capacity(T-AOC),sodium-potassium ATP enzyme activity(Na+-K+-ATPase)and calcium-magnesium ATP enzyme activity(Ca2+-Mg2+-ATPase)in the liver were measured,and the results of four biochemical indexes were compared.The correlation between Pearson and the bioluminescence parameters of intragastric administration of mice was analyzed.Results:1.The cold and hot properties of traditional Chinese medicine(Angelica sinensis,aconite,turmeric,perilla stem,fairy grass,Scutellaria baicalensis,honeysuckle vine,gardenia jasminoides,Artemisia annua,Fructus Corydalis)were determined by Scenic algae biological indicator method.The determined results are consistent with the previous measurements in our laboratory.20 traditional Chinese medicine(Gaoliang ginger,Cistanche deserticola,Wei Lingxian,longan meat,Corydalis,Chinese black,rose,maple skin,calendula,Millennium Health,Anemarrhena,Tianmendong,Radix Isatidis,plantain seed,semen Lepidii.,Rubiao,Aristolochia lanceolata,Platycladus orientalis leaves,Gentiana),among them,5 heat drugs(alpinia officinalis,Cistanche deserticola,Wei Lingxian,longan meat,Corydalis),6 cold drugs(Anemarrhena asphodeloides,Tianmendong,Radix Isatidis,plantain seed,Draba).The results of repeated determination are consistent with the results of previous measurements in our laboratory.It is still unable to successfully identify the medicinal properties of traditional Chinese medicine;the repeated determination results of the other nine drugs are either different from the previous determination results(consistent with the medicinal properties,it is considered that the identification is successful),or there are fluctuations,and so on,so they are removed in the subsequent mouse experiment.At the same time,Aristolochia manshuriensis has a strong hepatorenal toxicity,but also give up.The results show that the detection of cold and hot properties of traditional Chinese medicine based on ultra-weak luminescence of Scenedesmus has a certain degree of stability and repeatability.2.In the experiment of determining the ultra-weak luminous detection of mice to study the medicinal properties of traditional Chinese medicine,a series of basic conditions,such as body weight,week age,hair cover,drug concentration and dose,anesthetic concentration and dose,dark adaptation time,measurement time and so on,were determined in KM mice.On this basis,the best time of intragastric administration and detection time of mice were determined to be 6-10 days.At the same time,through the data collation and analysis,it was found that there was a statistical difference in the ratio of abdominal to back spontaneous luminous intensity between the gastric perfusion group and the blank group.10 kinds of traditional Chinese medicine(5 heat drugs,5 cold drugs)and 10 Chinese herbs(5 heat drugs,5 cold drugs)which have been successfully identified were used for detection after intragastric administration of 10 traditional Chinese medicine(5 heat drugs,5 cold drugs)and 10 Chinese herbs(5 heat drugs,5 cold drugs)after intragastric administration of 10 herbs(5 heat drugs,5 cold drugs).It was found that no matter whether the medicinal properties could be identified successfully by ultra-weak light emission detection,the abdominal back ratio of mice in the hot drug group was higher than that in the blank control group,and that in the cold drug group was lower than that in the blank control group.3.Through the measurement of the biochemical indexes of the liver of mice,it was found that among the four groups of biochemical indexes,there was a tendency that the biochemical indexes in the febrile drug group were higher than those in the cold drug group.In the correlation analysis,Except for a few drug groups(such as alpinia officinalis and Rhizoma Corydalis in T-AOC,aconite,longan meat,Rhizoma Anemarrhenae,plantain seed in SOD,perilla stem and gardenia in Na+-K+-ATP enzyme activity,There was no linear correlation between the biochemical indexes of Scutellaria baicalensis in the activity of Ca2+-Mg2+-ATP and the ratio of spontaneous luminous intensity of abdominal back(Scutellaria baicalensis).There was a significant correlation between the biochemical indexes and the ratio of ventral and dorsal SE intensity in other drug groups.The results showed that the change of metabolic level of mice induced by intragastric administration of traditional Chinese medicine showed the change of ultra-weak photon radiation,which proved that the experiment based on ultra-weak luminescence of mice had a certain degree of credibility.Conclusions:The experimental results of ultra-weak luminescence in intragastric administration of mice were consistent with the medicinal properties of traditional Chinese medicine contained in Pharmacopoeia,basically consistent with the results shown by biochemical indexes,and correlated with each other.These two methods have certain synchronization and integrity,which are related to the metabolic level of mice,and the results are the same as those recorded in Pharmacopoeia,and the ultra-weak luminous detection method has the advantages of non-invasive and easy to operate.According to the correct point of view,the ultra-weak luminous detection of gastric perfusion mice,like the method of biochemical indexes,can only roughly distinguish the cold and heat properties of traditional Chinese medicine,but can not be used as a method to distinguish the degree of cold and heat.In contrast,the operation of the Scenedesmus biological indicator method for detecting the medicinal properties of traditional Chinese medicine is simpler,the cost is lower,the results are more stable,the repeatability is better,and the degree of differentiation is higher.However,at present,it is still difficult to distinguish the cold and hot properties of a small number of traditional Chinese medicines.This may be related to the origin,batch,harvest season,processing methods and so on,and a large number of repetitive and in-depth studies need to be carried out. |
