Objective In this paper,we studied the quality standard of wheat bran as traditional Chinese medicine processing excipient by literature mining and the results of literature research.The study from the traits,microscopic identification,inspection,content determination and fingerprint,to establish specific quality standard method,optimize the process conditions of main quality components,supplemente and improve existing standards,to develop a more perfect wheat bran reliable quality standards.Methods Based on the clue of Li Dai Zhongyao Paozhi Zi Liao Ji Yao,backtracking and verifying the herbal works;and makes a literature survey on the Chinese Pharmacopoeia,the Processing Standards of Traditional Chinese Medicine of national and local and various databases.The use of eye,hand touch,smell,taste and other sensory methods of untreated wheat bran to observe and judge.The micro-structure of herbs can be identified by high power microscope.According to the Chinese Pharmacopoeia of 2015 edition of contained methods of determination of water,total ash and extract content,and specifies its limits.The basic chemical constituents of alanine,leucine and valine were used as the reference substance,and the best TLC conditions were explored to establish a simple and rapid TLC method.UV method was used to determine the content of wheat bran in flavonoids as the reference substance and HPLC method was used to determine the content of wheat bran in ferulic acid as the reference substance.The measured results were statistically calculated and the content limit was specified.According to the Chinese Pharmacopoeia of 2015 edition of contained methods of determination of aflatoxin B1,B2,G1,G2 and Organochlorine pesticide residue,and specifies its limits.The IR fingerprints of ten batches wheat bran samples from different areas were determined,and the similarity calculation and chemical pattern recognition were carried out.The effects of different packaging,ambient temperature and ambient humidity on the quality of wheat bran storage were investigated.Results Wheat bran as herbal medicine was first seen in the "Compendium of Materia Medica" in the Ming Dynasty and it was first seen as Traditional Chinese Medicine processing excipient in the Hua shi zhong cang jing.It has developed since the dynasties.There were more than 70 herbal medicines processed by Wheat bran before and in Qing Dynasty,among which 14 kinds of bran products are used,such as atractylodes,atractylodes,and tangerine peel.Nowadays,three kinds of excipients such as clear bran,honey bran and sugar bran are used.The collection of 30 batches of Wheat bran is divided by color,shape,gas,taste four aspects of trait description,can objectively reflect the quality characteristics of Pieces.The characteristics of starch grains,non-glandular hair,pericarp epidermis cells,cross cell and nucellus were observed under microscope,which was of great significance for the rapid identification of Pieces.The limits of 13.0%of the water content,6.0%of total ash content,and 14.0%of extract content of the 30 batches of Wheat bran are complementary to the quality of processing excipient.alanine,leucine and valine of Wheat bran have been identified by using TLC.Just as standardproducts,the simples showed the same stripe at the same location on the silica gel plate by using above method.The content of total flavonoids in Wheat bran have been determinated by UV method.It showed that the average content of total flavonoids was 0.58%in 30 batch of simples.Formulating containing rutin as total flavonoids shall not be less than 0.45%in the dried Wheat bran.The content of ferulic acid in Wheat bran have been determinated by HPLC method.It showed that the average content of ferulic acid was 9.15?g/g in 30 batch of simples.Combined with the actual situation,formulating containing ferulic acid shall not be less than 5.0?g/g in the dried Wheat bran.Aflatoxin B1,B2,G1,G2 and organochlorine pesticides BHC?DHT?PCNB were not detected in 30 batches of wheat bran samples.The fingerprint of wheat bran was established by infrared chromatography.It was found that 10 batches of wheat bran from different places had similar chemical composition,and there were obvious characteristic peaks at 3350,2929,1629,1542,1157,1020,575cm"1.It can be used as a basis for qualitative identification of wheat bran.Conclusion Based on the results of literature research,the quality standard of wheat bran was determined,and the traits,identification,inspection,content determination and Characteristic spectrum of wheat bran were studied in depth.Comparing the standard of wheat bran contained in the local standard,the trait standard has been improved and supplemented,the quality indicators on behalf of Wheat bran has been increased.The content control of TLC identification of TLC optimization results verify the superiority of the local standards of quality control.In the inspection items,moisture,total ash,and extractives were set as quality control limits,and aflatoxin and pesticide residues were examined.In the end,it is proposed to establish a more practical quality standard(draft)for the processing of wheat bran from Chinese herbal medicines.
