Objectives: Cold coagulation and blood stasis syndrome is one of the common TCM syndromes in cold areas.When cold pathogens invade blood,it is easy to damage Yang Qi,resulting in poor operation of Qi and blood,blood stasis stopped,thus forming cold coagulation and blood stasis syndrome.Cold coagulation and blood stasis syndrome is closely related to a variety of diseases,including internal,external,gynecological,pediatric,tumour and other diseases,which seriously threatens people's lives and health.Traditional Chinese medicine has certain advantages in the treatment of this syndrome.Doctors of past dynasties have also done a lot of work on this syndrome.In clinic,the treatment methods of this syndrome are mostly combined with warming yang,promoting blood circulation,promoting Qi or three branches,but there is little research on the weight of the three methods.The purpose of this study is to explore the key syndrome elements of cold coagulation and blood stasis syndrome,in order to provide accurate basis for clinical treatment of diseases.Methods: 1.The rat model of cold coagulation and blood stasis syndrome was established by freezing whole body in ice water.2.SD rats were randomly divided into pathological model group.The administration group included(aconite group,aconite Angelica group,aconite Angelica group,aconite Angelica group,aconite Angelica group,aconite Angelica group,aconite Angelica group,aconite group).Normal group was established.Rats in the administration group were given intragastric administration.The rats' symptoms,water consumption,heart rate and activity were observed.Blood was collected through abdominal aorta to detect differences.Whole blood viscosity at shear rate.3.The contents of TNF-alpha,interleukin-1beta,interleukin-6 and interleukin-10 in serum were detected by ELISA to observe the effects of different treatment methods on rats with cold coagulation and blood stasis syndrome.Result:1.After 15 days of administration,the water intake and heart rate of rats in each groupwere higher than those in the pathological model group in terms of activity,heart rate and water intake of Wenyang method(aconite group)and Wenyang Huoxue method(aconite group).The improvement of Wenyang method(aconite group)was the most obvious(P<0.01).2.The results of whole blood viscoity showed that WBV in pathological model group was significantly higher than that in normal group at all shear rates.It shows that cold coagulation can increase the blood viscosity of animals,thus forming blood stasis syndrome.At 10/S shear rate and 100/S shear rate,WBV of normal group,warm-yang method(aconite)and warm-yang method(angelica sinensis)were lower than pathological model group(P <0.001);at 180/S shear rate,WBV of warm-yang method(aconite sinensis)and warm-yang method(angelica sinensis sinensis)were lower than pathological model group(P < 0.01,P <0.05).The WBV of the pathological model group was higher than that of the normal group at each shear rate,P > 0.05,and there was no statistical significance.3.Effect of different treatment methods on TNF-alpha in rats with cold coagulation and blood stasis syndromeThe levels of TNF-a in serum of rats in Wenyang method(aconite),Wenyang Huoxue method(aconite sinensis),Wenyang Xingqi method(aconite sinensis)and normal group were lower than those in pathological model group(P < 0.001).Among them,Wenyang method(aconite sinensis)was lower than that in normal group(P > 0.05),Wenyang Xingqi method(aconite sinensis sinensis sinensis)was higher than that in normal group(P < 0.05),and the other five groups were lower than that in normal group(P < 0.001).4.Effect of different treatment methods on IL-1beta in rats with cold coagulation and blood stasis syndromeThe levels of IL-1beta in serum of rats treated by warming yang,activating blood circulation,promoting qi and combined therapy and normal group were lower than those of pathological model group.5.Effect of Different Therapies on IL-6 in Rats with Cold Coagulation and Blood Stasis SyndromeThe levels of IL-6 in serum of rats treated by warming yang,activating blood circulation,promoting qi and combined therapy and normal group were lower than those in pathological model group(P < 0.001).The levels of IL-6 in rats treated by Warming Yang(Fuzi group)were lower than those in normal group(P > 0.05),while those in other five groups were lower than those in normal group(P < 0.001).6.Effect of Different Therapies on IL-10 in Rats with Cold Coagulation and Blood Stasis SyndromeThe levels of IL-10 in serum of rats treated with warming yang,promoting blood circulation and promoting Qi were higher than those of rats treated with pathological model group(P < 0.001).The levels of IL-10 in serum of rats treated with Warming Yang method were higher than those of normal group(P > 0.05),and those in other groups were higher than those of normal group(P < 0.001).Conclusion:1.Warming Yang method can improve the symptoms of cold coagulation and blood stasis syndrome in rats,improve the heart rate,water consumption and activity,reduce the whole blood viscosity,improve microcirculation disturbance of cold coagulation and blood stasis syndrome,and thus improve local blood circulation.2.Wenyang method can reduce the content of TNF-alpha in serum,reduce platelet activating factor,reduce the content of IL-6 in serum,inhibit inflammatory reaction,increase the content of IL-10 inflammatory factor in serum,and improve the immunity of the body.3.Wenyang single method may replace the effect of three methods.4.Yang deficiency is the key factor of cold coagulation and blood stasis syndrome.
