| Objective and significance:Epilepsy,a functional disturbance of the CNS and induced by abnormal electrical discharge, manifestes by recurrent seizure.According to the different causes, epilepsy is generally divided into two categories:Idiopathic epilepsy and secondary epilepsy. The typical Clinical characteristics of epilepsy are:it happens recurrently, with unpredictable effects, and it needs long-term medication. Epilepsy is one of the most common diseases of nervous system, and it is the third most common neurological disease at home and abroad after cerebrovascular disease and dementia. According to the epidemiological data statistics, about50000000of the world population affected by this disease. Recent epidemiological studies in China show that, there are about9000000patients who have epilepsies. In China, the prevalence rate of epilepsy ranged from3.6‰-7‰. In the vast rural areas in China, the prevalence of epilepsy is about4.7‰-8.5‰, which is approximately equal to the whole world overall prevalence rate. Among them, about70%-80%of epilepsy patients did not receive treatment or received treatment which is not standard, and the mortality is about7.9/10million. Thus, the prevalence of epilepsy in China is close to that of developed countries, but the mortality rate is far higher than the world level, so this social phenomenon must be given adequate attention. All the time, epilepsy patients with normal population compared with, is thought to be a probability of cognitive and behavioral barriers to greater formation. Although there is an obvious individual differences, each epileptic patient still has cognitive ability damage risk. In patients with seizure frequency high, will damage the cognitive and behavioral capabilities continue to accumulate, so that their language ability, learning and memory, attention, orientation, behavior and emotions have different degrees of change.Nowadays, domestic and foreign treatment of epilepsy is the standard antiepileptic drugs, and the application of antiepileptic drugs needed to reach the goal is to eliminate seizures, but there is no effect on the pathological changes caused by epilepsy, and it also can’t block the process of epilepsy. Antiepileptic drugs almost all have more than a side effect, possibility and almost all antiepileptic drugs have cognitive impairment. Previous study showed that, Chinese medicine treatment of epilepsy has a long history, and has a far-reaching influence on the modern medicine and the posterity. Many classical prescriptions in still have important guiding significance to modern research, at the same time, traditional Chinese medicine treatment of epilepsy is also advancing with the times development. Compared with western medicine, traditional Chinese medicine has the advantage of low toxic side effects and low price.The maximal electroshock seizure model (MES), Pentetrazole (PTZ) seizures model, Strychnine (STR) seizures model and Pilocarpine (PLCP) seizures model is the most widely used to build the seizure model. The durg which can control the seizure of the mice on these four models, is considered that may control the clinical seizures.Our preliminary study showed that bupleurum saponins in MESã€PTZã€STR model has anticonvulsive action. Other research shows:radix scutellariae in PLCP epilepsy model has anticonvulsants and nerve protective effect. Medicine pair is the most basic form, in the development of traditional Chinese medicine has a long history, many classic medicine pair is used till now. For modern Chinese herbal medicine for clinical research, theoretical research and experimental research have different degree of progress and development.The combination of medicine, is not only to do a simple addition, between the elements of the consequent is usually the complicated chemical and physical changes, including the chemical composition will be changed. Research also suggests that can improve the solubility of compatibility of medicine to improve drug targeting and pharmacokinetic behavior change.Zhang Zhongjing used the combination of radix bupleuri and radix scutellariae to treat jaundice, liver depression and qi stagnation due to the new product mass and so on the many kinds of disease, the effect of that is remarkable. Compendium of materia medica says,"Radix bupleuri brothers shaoyang, Radix scutellariae is to assist.Ben Cao Shu Gou Yuan says about radix bupleuri "XuanChang veins,is better to use radix scutellariae together". Our preliminary advocated epilepsy treated from liver, Chai Hu Shu Gan Tang has achieved a satisfactory therapeutic effect in clinic. Radix bupleuri as monarch drug in the party, with radix scutellariae altogether plays compromise shaoyang, discussion on the liver function, but also hydrophobic factors combined with nourishing liver liver. So we established the acute MESã€PTZã€STR〠PLCP model, to further explore the antiepileptic effect of radix bupleuri radix scutellariae compatibility application.Method and content1Building and processing the MES modelThe required mouse were prepared by screening through the pharmacological multi-functional implemen.60KM mouse of SPF grade were prepared, which were then divided into6groups. The group A (Model Group) was given equal volume of physiological saline; the B and C were given the dosage in which proportion of radix bupleuri and radix scutellariae are5-2and1-1respectively, and of which the concentration of radix bupleuri are0.3g/ml and0.21g/ml respectively; the D was given radix bupleuri; the E was given radix scutellariae, of which the concentration is0.42g/ml; the F was given diazepam.Atter, of which the concentration is0.4g/ml. After1hours of the last administration, electric stimulation were given, the were recorded, and the were calculated.2Making of chronic PTZ kindling rat model60KM mouse of SPF grade were prepared. The grouping and the administration methods are the same as MES mouse model. After1hours of the last administration, all groups of rats were injected with85mg/kg pentetrazole by peritoneal, then the30-min epileptic seizure were observed immediately, the classification of which seizure was in accordance with the Racine standards. For the next step, the incubation period of epileptic seizure were recorded, including the latency of clonic convulsion(Since the injection of pentetrazole till time of attack grade1to grade3)〠the latency of tetanic convulsion (Since the injection of pentetrazole till the time of attack grade5)ã€and the incubation period of death.3Building and processing chronic STR kindling rat model60KM mouse of SPF grade were prepared. The grouping and the administration method are the same as MES mouse model. After1hours of the last administration, all groups of rats were injected with2mg/kg vauqueline by peritoneal, then the15-min epileptic seizure were observed immediately and the incubation period clonus incubation period and death of the mouse were recorded.4Building and processing the chronic PLCP kindling rat model60KM mouse of SPF grade were prepared. The grouping and the administration method are the same as MES mouse model. After1hours of the last administration, all groups of rats were injected with400mg/kg Pilocarpine by peritoneal, then the30-min epileptic seizure were observed immediately and the incubation period clonus incubation period and death of the mouse were recorded.5Statistical methodsThe statistical analysis of the data was conducted by the SPSS13.0statistics software. The measurement data was indicated by the mean value and standard deviation (x±s),and the result comparison of each group was conducted by the Repeater Measure variance analysis, the multiple comparisons by the Tukey law, and the rate comparison of tetanic convulsion by R×C Chi-Square Test; P<0.05represents that the difference indicates statistical significance. P<0.01represents that the difference indicates remarkable statistical significance. Result1The effect of Bupleurum and Radix Scutellariae application of MES model.The statistical significance of the convulsion rate between groups(χ2=59.000, P <0.001), Group Bã€Group Cã€Group D and Group F could reduce the rate of seizures in mice, and the most significant decrease appear in Group B and Group F.2The effect of the application of Bupleurum and Radix Scutellariae on the chronic PTZ kindling rat model during the onset of clonic latency,the seizure latency and the death latency.Group B, C, D, E, and F can extend the onset of clonic latency the latency of seizure and the latency of dean. Compared with Group A, GroupC, D and E do not have significant differences. However, Group B and F have significant differences.3The effect of the application of Bupleurum and Radix Scutellariae on the chronic STR kindling rat model during the onset of clonic latency,the seizure latency and the death latency.Group B,C,D,E and F can extend the onset of clonic latency. However, compared with group there is no statistical significance. Group B, C, D, E and F can extend the death latency. Compared with group A, Group C, D and E have no statistical significance, but Group B and F have (P<0.01). Also compared with Group C, D and E, Group B has statistical significance.4The Effect of the application of Bupleurum and Radix Scutellariae on the chronic PLCP kindling rat model during the during the onset of clonic latency and the death latency.Group B, C, D, E and F can extend both the onset of clonic latency and the death latency. However, compared with Group A, there is no statistical significance except F (P<0.01). ConclusionThe compatibility of radix bupleuri and radix scutellariae are anticonvulsant, antiepileptic role,the effect of that group is evident,which the ratio of radix bupleuri and radix scutellariae is5-2. |
