Dynamic Mechanism Study Of Metabolic Response In Peptic Ulcer Rats With Electro-acupuncture Treatment

AIM:To study the effect of electro-acupuncture at gastric meridians on the repair of gastric mucosal lesions in rats with peptic ulcer,and the metabolic response mechanism and dynamic mechanism of gastric mucosal,liver and kidney tissue samples by 1H-NMR metabonomic analysis of gastric mucosal,liver and kidney tissue samples,combined with morphological and histopathological observation of gastric mucosa.Methods:54 male clean-grade rats were randomly divided into normal group(N group),model group(M group)and electro-acupuncture group(EA group)according to random number table method.Each group was further divided into three subgroups according to different intervening time,namely 1-day subgroup,4-day subgroup and 7-day subgroup.Except for the normal group,rats in other groups were given 70%ethanol to induce peptic ulcer model.After the model was established,rats in the electro-acupuncture group were given electro-acupuncture at Zusanli and Liangmen acupoints.After the intervention,gastric mucosal tissues were taken to observe the gross morphology of the naked eye and histopathological changes under light microscopy.The expression of metabolites in stomach mucosal,liver and kidney and their dynamic changes were detected by the NMR metabolic group.Results:Histopathological observation of gastric mucosal showed that the epithelium of gastric mucosa in normal group was intact,the glands arranged closely,the shape and size were normal,and there was no obvious congestion and edema.In model group,the glands arranged disorderly,most glands were destroyed,and a large number of inflammatory cells infiltrated in the glands.There were obvious congestion and edema.However,on the 7th day,gastric mucosal pathology improved.In EA1 group,the gastric mucosa was still damaged.In 4-day group and 7-day group,the gastric mucosa was repaired.1H-NMR results showed that 22 different metabolites were found in rat stomach mucosal,liver and kidney tissues,which were related to 18 metabolic pathways:aminoacyl-Tma biosynthesis;valine,leucine and isoleucine biosynthesis;phenylalanine,tyrosine and tryptophan biosynthesis;D-glutamine and D-glutamine metabolism;glycine,serine and threonine generation.Metabolism of alanine,aspartic acid and glutamic acid;phenylalanine metabolism;methane metabolism;taurine and hypoxanthine metabolism;arginine and proline metabolism;glycerin metabolism;glycerophosphate metabolism;glutathione metabolism;cysteine and methionine metabolism;inositol phosphoric acid metabolism;tyrosine metabolism;primary bile acid biosynthesis;tricarboxylic acid cycle(TCA cycle).According to the physiological functions,the related metabolites in these metabolic pathways can be divided into neurotransmitter-related metabolism,energy(ATP)related metabolism,cell and cell membrane metabolism and other biosynthesis-related metabolism.Conclusion:Electro-acupuncture can effectively treat peptic ulcer.The therapeutic effect is not obvious after one day of treatment.After four days of treatment,it can effectively reverse gastric mucosal injury caused by peptic ulcer.After seven days of treatment,the therapeutic effect tends to be stable.Peptic ulcer can affect neurotransmitters,energy cells,antioxidants,tissue repair and other related metabolic pathways.Electro-acupuncture for 4 days can significantly reverse the concentration of related metabolites.The effect of electro-acupuncture for 7 days tends to be stable.Therefore,this study showed that electro-acupuncture treatment of peptic ulcer 4 days the most obvious effect.The molecular response mechanism and further analysis of metabolites still need to be further studied.
