| Objective: This paper aims to experimentally observe the effects of the Chinese herbal medicine of tonifying kidney and harmonizing qi and blood on the uterine artery blood flow,VEGF and s Flt-1 levels in the serum of blood-stasis induced abortion rats in early pregnancy.In addition,the effects and mechanism of the Chinese herbal medicine of tonifying kidney and harmonizing qi and blood in improving the uterine blood flow perfusion and decreasing the abortion rate are explored,therefore serving an objective basis for clinical application.Methods: After adaptive feeding,healthy female adult SPF rats were mated.Among which,the pregnants were randomly divided into six groups: the normal pregnancy group(the group A),the model group(the group B),the low dose group of TCM(the group C),the medium dose group of TCM(the group D),the high dose group of TCM(the group E)and the aspirin group(the group F),with 10 rats in each group.The group A was set as control,and the remaining five groups were experimental.Methods: All rats in experimental groups were injected with epinephrine hydrochloride 0.9 mg/kg subcutaneously once a day from the 1st day of pregnancy until the 11 th to form blood stasis.Mifepristone solution 3.75 mg/kg was given intragastrically on the 8th day of pregnancy to induce abortion.Meanwhile,each experimental group was treated respectively for 11 days from the 1st day of pregnancy: the group A and B were given distilled water 10 m L/kg;the group C,D and E were given Chinese herbal decoction6.525g/kg,13.05g/kg and 26.1g/kg respectively;and the group F was given aspirin solution 6.75 mg/kg.On the 12 th day,all groups were anesthetized by intraperitoneal injection of chloral hydrate in order to measure uterine artery blood flow parameters under the Color Doppler Ultrasonography.Then the abdominal cavities were opened and the numbers of implantation points,normal embryos and abortions were counted and recorded;the abortion rate was thus calculated.Lastly,the abdominal aorta blood was collected and the serum was obtained after centrifugation.And the VEGF and s Flt-1 levels in the serum were measured by ELISA.Results:(1)The abortion number and abortion rate: Compared with the group A,the abortion number and abortion rate of the group B were very significantly increased(P<0.01).Compared with the group B,the abortion number of the group C,D and E were very significantly reduced(P<0.01),while there was no significant difference between the group F and the group B(P > 0.05).In abortion rate,the group C,D,E and F were significantly reduced(P<0.05 or P<0.01).Compared with the group F,the abortion number and the abortion rate in the group C,D and E were very significantly different(P<0.01).(2)PI,RI,S/D of uterine artery: Compared with the group A,PI,RI and S/D of uterine artery in the group B were very significantly increased(P<0.01).Compared with the group B,PI,RI and S/D in the group C,D,E and F were very significantly decreased(P<0.01).Compared with the group F,PI,RI and S/D of the group E were significantly decreased(P < 0.05),while the group B and C were no statistical difference(P>0.05).(3)VEGF levels in the serum: Compared with the group A,the VEGF level in the serum in the group B was very significantly lower(P<0.01).Compared with the group B,the VEGF level in the serum in the group C,D and E were significantly higher(P<0.05 or P<0.01),and that in the group F was not significantly different(P>0.05).Compared with the group F,the group D and E increased very significantly(P<0.01),while the group C had no statistical difference(P>0.05).(4)s Flt-1 levels in the serum: Compared with the group A,the s Flt-1 level in the serum in the group B was very significantly higher(P<0.01).Compared with the group B,the serum s Flt-1 level in the group C,D and E were very significantly lower(P<0.01),while there was no significant difference in the group F(P>0.05).Compared with the group F,the serum s Flt-1 level in the group C,D and E were very significantly lower(P<0.01).Conclusions:(1)Chinese herbal medicine of tonifying kidney and harmonizing qi and blood can improve uterine artery blood perfusion and reduce abortion rate in abortion rats caused by blood stasis,thus showing this method has positive effect on fetal protection in abortion rats caused by blood stasis in early pregnancy.(2)Chinese herbal medicine of tonifying kidney and harmonizing qi and blood may promote uterine vascular recasting by increasing the VEGF level and reducing the s Flt-1 level,therefore reduces the resistance of uterine artery blood flow and improves the uterine blood perfusion,and ultimately increases the success rate of pregnancy. |
