Objective:Detecting the expression of microsatellite instability(MSI),vascular endothelial growth factor(VEGF)and membrane-type6 matrix metalloproteinase(MT6-MMP)in gastric cancer(GC)tissues,to explore their correlation with the clinicopathological features and postoperative peritoneal metastasis of GC.And to analyze the relationship between the expression of MSI,VEGF and MT6-MMP.Methods:60 cases of tissue sample of gastric adenocarcinoma who were resected and diagnosed in the First Affiliated Hospital of Yangtze University from 2014 to 2016were collected.42 cases of GC with postoperative peritoneal metastasis and 18 cases of GC without postoperative peritoneal metastasis during 2 years of follow-up.Immunohistochemistry(IHC)streptavidin perosidase(SP)method was used to detect the expression of MSI,VEGF and MT6-MMP in GC tissues,and then used the SPSS19.0 statistical software to analyze the experimental results,and P<0.05 was considered statistically significant.Results:1.The MSI was not related to patient,s gender(?~2=1.374,P=0.241),age(?~2=3.857,P=0.050),location(?~2=3.859,P=0.088)and diameter(?~2=0.267,P=0.605),but related to differentiation degree(?~2=19.543,P=0.000),clinical stage(?~2=6.023,P=0.014)and lymph node metastasis(?~2=7.972,P=0.005).Compared with MSI-H group,MSI-L/MSS group had lower differentiation degree,later clinical stage and higher lymph node metastasis rate.2.The VEGF was not related to patient,s gender(?~2=0.002,P=0.967),age(?~2=0.011,P=0.918),location(?~2=0.019,P=0.961)and diameter(?~2=0.020,P=0.887),but related to differentiation degree(?~2=9.627,P=0.002),clinical stage(?~2=5.293,P=0.021)and lymph node metastasis(?~2=14.264,P=0.000).Compared with the low expression group,the high expression group had lower differentiation degree,later clinical stage and higher lymph node metastasis rate.3.MT6-MMP was not related to patient,s gender(?~2=0.000,P=1.000),age(?~2=0.000,P=1.000),location(?~2=0.278,P=0.852)and diameter(?~2=0.208,P=0.648),but related to differentiation degree(?~2=14.944,P=0.000),clinical stage(?~2=4.904,P=0.027)and lymph node metastasis(?~2=9.627,P=0.000).Compared with the low expression group,the high expression group had lower differentiation degree,later clinical stage and higher lymph node metastasis rate.4.The high expression rates of MSI,VEGF and MT6-MMP in GC with postoperative peritoneal metastasis were 9.5%,95.2%,97.6%respectively,the high expression rates of MSI,VEGF and MT6-MMP in GC without postoperative peritoneal metastasis were and 44.4%,44.4%and 33.3%respectively.There were statistical differences between the two groups(?~2=7.545,P=0.004;?~2=23.642,P=0.000;?~2=20.376,P=0.000).The peritoneal metastasis rate in MSI-L/MSS group,VEGF-high expression group and MT6-MMP-high expression group was higher than MSI-H group,VEGF-low expression group and MT6-MMP-low expression group.5.MSI was negatively correlated with the expression of VEGF in GC tissues(r_s=-0.625,P=0.000),also negatively correlated with the expression of MT6-MMP(r_s=-0.611,P=0.000).The VEGF was positively correlated with the expression of MT6-MMP in GC tissues(r_s=0.451,P=0.000).Conclusion:MSI,VEGF and MT6-MMP are closely related to the pathological characteristics of GC and postoperative peritoneal metastasis,and the expression levels of three are correlated.They can be used as high risk factors to evaluate the progression of GC,they have important reference value in predicting peritoneal metastasis and guiding individualized treatment.They are expected to be used as molecular markers to predict the prognosis of GC.
