Objective:To provide preliminary information for ovarian cancer angiography fundamental research,preparing nano phase change contrast agent(NPCT)carrying HE4 for ovarian cancer and to investigate its basic characteristics and observe its enhanced ultrasound imaging effect in vitro.Methods:1.The PFH-PLGA nanopolymer contrast agent was prepared by single emulsification from PLGA-COOH and PFH.2.HE4-PEG-NH2 was dispersed and dissolved in MES buffer solution by carbodiimide method,and then mixed with the contrast agent obtained after centrifugation.After incubating at room temperature,washing with double steaming water and centrifuging,HE4-PFH-PLGA nanopolymer contrast agent was obtained.3.The size,morphology and surface potential of HE4-PFH-PLGA nanopolymer contrast agent were measured in vitro.4.The binding rates of PFH-PLGA nanopolymer contrast agent and HE4-PFH-PLGA nanopolymer contrast agent to the second antibody were detected by flow cytometry.5.The phase transition of HE4-PFH-PLGA nanopolymer contrast agent was observed by high intensity focused ultrasound(HIFU)stimulation in vitro.Results:1.The HE4-PFH-PLGA nanopolymer contrast agent has a milky white suspension,regular shape and spherical capsule.The particle size is 312-523 nm,concentrated at 406 nm,concentration is 1*106/ml,and the average surface potential is(-2.42+4.52)mV 2.The binding rates of the second antibody to PFH-PLGA nanopolymer contrast agent and HE4-PFH-PLGA nanopolymer contrast agent0.72%wereand 82.5%,respectively.3.When the temperature of HIFU was more than 70?in vitro,the echo declaration of HE4-PFH-PLGA nano-macromolecule contrast agent water sac was enhanced under the mode of fundamental wave(mechanical index 0.6).The echo in the water sac was dotted,small and uniform,and the echo in the rear was not attenuated significantly.In the control group,the echo in the double-steamed water sac was hypoechoic,and the echo declaration in the rear was enhanced.Conclusion:The HE4-PFH-PLGA nanopolymer contrast agent has small particle size and good stability.It can enhance ultrasound in vitro HIFU.
