Objective:To determine the content of adenosine in rat myocardial tissue by HPLC,and to investigate the effect of electroacupuncture pretreatment on myocardial ischemia reperfusion injury in rats.To investigate the effects of electroacupuncture pretreatment time(7 days and 14 days)on adenosine in myocardial tissue.Methods:1.80 male SD rats were randomly selected and divided into two groups for 7days and 14 days.The large group was divided into the normal group,the sham operation group,the MIRI model group,the electroacupuncture Neiguan(PC6)modeling group,and the electroacupuncture Hegu(LI4)modeling group,with 8 rats in each group.2.The normal group was raised normally to the specified days.The sham operation group was raised normally to the specified days,and the chest was opened on the same day.The MIRI model group was raised normally to a specified number of days.The MIRI rat model was replicated in the electroacupuncture Neiguan(PC6)and Hegu(LI4)groups,and the two groups were bound on the operating table for 30 min daily.Electrocardiogram test was performed during modeling.3.After sampling,the myocardial tissue was detected by High-performance liquid chromatography(HPLC)with Venusil MP C18 column(4.6mm×250mm,5um)(mobile phase: A:PH=5.8 phosphate buffer,and B was methanol).The detection wavelength was 254 nm,and the column temperature was 26.5??24??26.5?.The content of adenosine in each group was detected.Results:1.In the 7-days group,compared with the normal group,the levels of adenosine in the sham operation group,MIRI model group,electroacupuncture Neiguan(PC6)modeling group and electroacupuncture Hegu(LI4)modeling group all decreased,among which,the levels of adenosine in the sham operation group and MIRI model group decreased significantly(p<0.05,p<0.05).Compared with the MIRImodel group,the content of adenosine in the electroacupuncture Neiguan(PC6)modeling group and the electroacupuncture Hegu(LI4)modeling group were significantly increased(p<0.05,p<0.01),and the content of ATP,ADP and AMP in the electroacupuncture Hegu(LI4)group were significantly higher than that in the MIRI model group(p<0.01,p<0.01,p<0.01).In 14-days group,compared with the normal group,the levels of adenosine in the sham operation group,MIRI model group,electroacupuncture Neiguan(PC6)modeling group and electroacupuncture Hegu(LI4)modeling group all decreased.Compared with the MIRI model group,the content of adenosine in the electroacupuncture Neiguan(PC6)plus model group and electroacupuncture Hegu(LI4)plus model group were significantly increased(p<0.05,p<0.05),and the content of ATP and ADP in both groups were higher than that in the MIRI model group.2.In 7-days group,the content of ATP,ADP,AMP and adenosine in the group of electroacupuncture Hegu(LI4)plus modeling increased significantly(p<0.01,p<0.01,p<0.01)compared with that in the group of electroacupuncture Neiguan(PC6)plus modeling.However,in the 14-day group,the content of ATP in the electroacupuncture Hegu(LI4)plus modeling group increased significantly,and there was no significant statistical difference in the content of ADP,AMP and adenosine compared with that in the electroacupuncture Neiguan(PC6)plus modeling group.3.Compared with the 7-day group,the contents of ATP,ADP,AMP and adenosine in each group in the 14-day group increased significantly,showing statistical significance.Conclusion:1.Electroacupuncture Neiguan(PC6)l preconditioning and electroacupuncture Hegu(LI4)preconditioning can alleviate the decrease of ATP,ADP and adenosine induced by myocardial ischemia-reperfusion injury,and enhance the anti-hypotrophic ability of myocardial tissue.The protective mechanism may be related to the increase of adenosine content in energy metabolism,which plays a role in dilating blood vessels and protecting cardiac tissue.2.The specificity of electroacupuncture Hegu(LI4)pretreatment in alleviating myocardial ischemia reperfusion injury is better than that of electroacupuncture pretreatment Neiguan(PC6)3.There was no difference in electroacupuncture pretreatment for 14 days as compared with electroacupuncture pretreatment for 7 days.
