The traditional prescription Shengyang Yiwei Decoction originated from“ Treatise on Spleen and Stomach ” written by Lidongyuan,a famous medical expert in Jin-Yuan Dynasty of China.It has been used broadly in clinic for a long time due to the remarkable effects on invigorating vital energy,strengthening spleen,nourishing blood and regulating menstruation.Shengyang Yiwei Decoction was included in the catalogue of Ancient Classical Prescriptions(first batch)in April2018,which made the research on the new formulation of the decoction has been widely concerned.At the same time,the modernization process of Shengyang Yiwei Decoction has been greatly accelerated.In this study,The chemical research of Shengyang Yiwei Decoction was explored by means of historical textual research and experimental study.The research contents are mainly carried out in the following five aspects.1.Historical textual research of Shengyang Yiwei DecoctionThrough literature research,the historical textual research on "Shengyang Yiwei Decoction" was carried out,including the textual research on each herbal medicine of traditional Chinese medicine and weights and measures in Jin-Yuan Dynasty.As a result,the origins of 14 herbs in the prescription and the modern significance of 'liang','qian' and 'zhan' during the Jin-Yuan Dynastywere clarified.2.Quantitative research of adenine,uridine and guanosine in Pinellia ternata by HPLCThe water extractionof Pinellia ternata was analyzed and identified by using UPLC-Q-TOF-MS.The result showed that the nucleoside components are the main chemical component of Pinellia ternata.On this basis,a quantitive method of HPLC was developed for the determination of adenine,uridine and guanosine in Pinellia ternata.3.Study on the processing technologies of Pinellia ternata of Sheng yang YiweiDecoctionBy comparing tthe contents of calcium oxalate and nucleosides between Pinellia ternata and the processed products,the nucleosides(adenine,uridine,guanosine)were determined as the evaluation index of processed Pinellia ternata.The orthogonal experimental was used to investigate the water addition,the time of washing and the number of washing time in the processing of Pinellia ternata.Finally,the optimal processing technology was selected.4.The improvement of the quality standard for Atractylodes rhizomeBecause the standard of Atractylodes macrocephala in the Chinese Pharmacopoeia of the 2015 edition lacks content determination items of index components,A method for determination of Atractylenolide?and Atractylenolide? in Atractylodes rhizome by HPLC was established.The results of methodological evaluation showed that this method was reliable and stable,which could be applied for the quality control of Atractylenolide?and Atractylenolide ? in Atractylodes rhizome.5.Analysis of volatile oil in Shengyang Yiwei DecoctionThe composition and relative contents of volatile oil in Shengyang Yiwei Decoction.The result showed that there were many components of volatile oil in the raw medicine material of Shengyang Yiwei Decoction,mainly from Qianghuo,Duhuo,Chen Pei,Fangfeng and Bupleurum.However the water extraction of Shengyang Yiwei Decoction did not.6.Determination of the contents of index ingredients in Shengyang Yiwei DecoctionA method for the quantitation of berberine hydrochloride,calycosin-7-glu coside,hesperidin and glycyrrhizic acid in Shengyang Yiwei Decoction.The resu lts of methodological evaluation has shown that this method had a good precisio n,repeatability,reproducibility and recovery.It is suitable to determine the con tents ofthese 4 constituents in Shengyang Yiwei Decoction.In conclusion,this study takes Shengyang Yiwei Decoction as the research object.,It maily studied the historical textual of Shengyang Yiwei Decoction,the processing technology of Pinellia ternata,the improvement of the quality stan dard for Atractylodes rhizome,the analysis of volatile oil components and the q uantitative determination of index ingredients in Shengyang Yiwei Decoction.It provides a scientific basis for the preparation of new formulation of Shengyang Yiwei Decoction.
