At present, the quality control of pinellia ternate in 2010 edition of "Chinese Pharmacopoeia" was messured by the determination of total acid content by the potentiometric titration method, this method issensitive and fast, the quality of Pinellia can be reacted to a certain extent. But this component is complex and the index components of it are not clear, in recent years there search on establishing the index components of pinellia exploration and the quality standard was more and more important. The polar water soluble components of pinellia were studied in this paper, the main works are as follows:First, the section of literature review:The three sections are the the chemical constituents studies of pinellia separation, current situation of HPLC separation and the application of HILIC in the analysis of traditional Chinese Medicine.Second, the sections of experimental:1.The fingerprints of the Pinellia ternate and its processed products which is collected in the 2010 edition of "Chinese Pharmacopoeia" was established for the study of the water soluble components of them:a Waters X-Bridge C18 cloumn (4.6×150mm,5μm) was used, the mobile phase was acetonitrile (A)-0.05% Phosphoric acid (B) with gradient elution; the column temperature was 30 ℃; the flow rate was 1mL/min; the detection wavelength is 286nm. In this chapter the method of precision, reproducibility, sample stability were investigated, and the results were satisfactory, indicating that the precision of the method, reproducibility were well, and the samples remained stable within 20 hours.2.The HILIC fingerprints of the pinellia ternate was established for the study of the water soluble components of them:a Welch UltimateR XB-NH2 (4.6×250mm,5μm) was used, the mobile phase was acetonitrile (A)-water (B) with gradient elution; the column temperature was 30℃; the flow rate was 1mL/min; the detection wavelength is 265nm. And then the RP-fingerprints of pinellia ternate and the HILIC fingerprints of it were compared to explore the relevance between the components that were separated under the differert modes, The result can provide a basis for the more comprehensive, more accurate method for the Study of quality control of pinellia ternate.3.The polar alkaloid components was studied under off-line 2D-LC-MS which is established C18 cloumn× NH2/HILIC/C18-SCX cloumns so that we can get as much component informations as possible.
