Morinda officinalis How is the dry root of Morinda officinalis,which is one of the four major South medicines in China.It has the functions of tonifying kidney deficiency,strengthening bones and strengthening bones,treating rheumatic pain and relieving fatigue.At present,the research mainly focuses on the efficacy of crude extracts,and the research on functional ingredients is relatively insufficient.In order to strengthen the research on the deep processing and biological activity of Morinda officinalis,this research aims to investigate the anti-depressant and anti-fatigue active ingredients of Morinda officinalis How.1.Morinda officinalis How was repeatedly extracted twice by 75% ethanol,and concentrated under reduced pressure to obtain the ethanol extract of Morinda officinalis,the yield was 21.7%;The extraction was repeated twice by deionized water,concentrated under reduced pressure to obtain the extract of Morinda officinalis How,and then treated with 95% ethanol alcohol precipitation to obtain alcohol precipitation and alcohol solubles,the yields were 12.3% and 8.6%.Through the activity test of anti-depression and anti-fatigue mice behavioral models,the results show that the alcohol precipitation of Morinda officinalis has the most significant anti-depressant activity.In addition,Morinda officinalis ethanolic extracts has the most significant anti-depressant activity.2.The active constituents was respectively separated and purified by Hw-55 F and Hw-40 gel column from the alcohol precipitation of Morinda officinalis How.The structure of the obtained product was identified by NMR.Through the activity test of anti-depression mice behavioral models,the results show that the component D-2 has the most significant anti-depressant activity.By measuring the concentration of dopamine in the hippocampus and striatum of each group of mice,the DA concentration in the D-2 group was significantly higher than that in the blank group.After separation by ODS-AQ column,four compound monomers were further separated from Component D-2,which were identified by NMR: nystose(1),inulin-type pentasaccharide(2),inulin-type hexasaccharide(3),inulin-type heptasaccharide(4).3.The experimental group were treated with component D-2 in two ingestion methods: intragastric and intraperitoneal injection.The results showed that in the intragastric group,the component D-2 can play a role in anti-depressant activity.4.The active constituents was respectively separated and purified by macroporous resin,MCI,and ODS column chromatography from Morinda officinalis ethanolic extracts.The structure of the obtained product was identified by NMR.Through the activity test of anti-fatigue mice behavioral models,the results showed that the component E-1 has the most significant anti-depressant activity.After separation by ODS-A and ODS-AQ column,four compound monomers were further separated: 4-carboxyl-7-hydroxyl-8-hydroxymethyl-3,6-iridoidglycosides(5),4-acetyl-8-hydroxymethyl-3,7-iridoid-glycosides(6),4-carboxyl-7-hydroxyl-8-hydroxymethyl-10-acety l-3,6-iridoidglycosides(7).Compounds 5,6 and 7 are firstly isolated and identified from Morinda officinalis How,which will provide a theoretical basis for clarifying the exploitation value of the resource to be developed into healthcare products.
