| ObjectiveUnder the guidance of traditional Chinese medicine(TCM)theory,this project investigated Saururus chinensis(Lour.)Baill for its quality control.Based on the original quality standards,according to the technical requirements of the TCM new drug development,comprehensive analysis methods were adopted to carry out researches like raw material quality control,origins,harvesting and processing procedures,as a result,the optimum choice of the above-mentioned factors will be discovered,and a more comprehensive quality control method for Saururus chinensis(Lour.)Baill will be established.MethodOrigin survey(1)Field investigation:To investigate the distribution of wild resources of Saururus chinensis(Lour.)Baill.and evaluate their exploitation.The morphology,growth environment,growth cycle,threat of pests and diseases,and local use of Saururus chinensis(Lour.)Baill.were investigated,and the distribution and growth of Saururus chinensis(Lour.)Baill.were summarized.(2)Market research:Investigate the origin,principal use,processing method,yield,market price,etc.of the Saururus chinensis(Lour.)Baill.on the market,and summarize the information such as processing methods and yield.2.Study on quality standards of medicinal materials(1)Collect Saururus chinensis(Lour.)Baill.and test according to the standards of Saururus chinensis(Lour.)Baill.recorded in the 2015 edition of the Chinese Pharmacopoeia,and included the qualified medicines for further study.(2)The effective chemical constituents in Saururus chinensis(Lour.)Baill.were studied by ultra performance liquid chromatography-quadrupole-time-of-flight mass spectrometry(UPLC-Q-TOF).Through the matching with the database and the literature comparison analysis,the chemical structures found in the Saururus chinensis(Lour.)Baill.were clarified.(3)Research on fingerprints of medicinal materials.High performance liquid chromatography(HPLC)method was used to study the fingerprints of Saururus chinensis(Lour.)Baill.of different producing areas and different batches,The chromatographie conditions and sample preparation methods were optimized comprehensively,and the methodological investigation was completed.For the obtained chromatogram,The National Pharmacopoeia’s fingerprint similarity software was used to calculate the similarity,and combined with data analysis carried out by SPSS software.Then,Thin-layer chromatography(TLC)fingerprint for different producing areas and different batches were also collected.(4)Rapid identification method of TLC,using a variety of lignan compounds and various flavonoids as indicators to establish a raPid identification method for the thin layer chromarography of Saururus chinensis(Lour.)Baill.(5)The method for determination of total flavonoids and total lignans in Saururus chinensis(Lour.)Baill.was established by ultraviolet spectrophotometry(uv).Methodological investigations such as linear range,precision,repeatability,stability,and sample recovery were performed.For simultaneous determination of various lignans and flavonoids,Ultra-high performance liquid chromatography(uplc)was used.For such part of experiment,methodological studies such as specificity,linear range,precision,repeatability,stability,and sample recovery were performed.3.Evaluation of harvesting and processing technology and quality control of Saururus chinensis(Lour.)Baill.from different producing areasUsing the established quality standards of Saururus chinensis(Lour.)Baill.,the quality of different drying processes and different harvesting seasons was compared,and the optimal conditions were obtained,as well as the best origins.ResuIt1.Origin surveyThe results showed that Saururus chinensis(Lour.)Baill.is a small variety of traditional Chinese medicine,with a small amount of consumption,mainly wild resources,rich in resources,widely distributed in Hebei,Henan,Shandong and the Yangtze River basin and its south.This study based on the three districts of Duanfen Town,Taishan,Guangdong,Chongzuotian,Guangxi,and Luotian County,Huanggang,Hubei Province.The morphology,growth environment,growth cycle,pests and diseases,local use,main uses,processing methods and yields were investigated.The results showed that no significant differences was found in the quality caused by changes in plant status,growth environment,quality of medicinal materials,local application,processing of origin,processing,storage and transportation in producing area of Saururus chinensis(Lour.)Baill.,which were all wild wild areas near streams and ditches.In the side or wet sand,the distribution is scattered;affected seriously by pests,causing the resources decreasing;the local application is rare,mostly for fresh roots;the post-harvest processing of medicinal materials is mostly sun exposure;the storage environment is simple,and the temperature and humidity are not controlled and the herb was easily wetted.The three producing areas are harvest.ed in the Saururus chinensis(Lour.)Baill.in March-August,May/June,and in spring and summer of a year;the yields are also different,but they are all below 1000 kg,which is compared with the medicinal materials.Less relevant.Study on the quality standard of Saururus inB,5-(2,5-Dimethyl-Benzyloxy)-2-Methyl-Benzofuran-3-Carboxylic Acid Methyl Ester).(2)Fingerprint research:Established the HPLC fingerprint of Saururus chinensis(Lour.)Baill..A common pattern of HPLC fingerprints of Saururus chinensis(Lour.)Baill.was established,and 19 common peaks were identified,and 10 common peaks were identified.The 1th peak were rutin,the 2th peak was hyperin,and the 3th peak was quercitrin,the 5th peak is aristolactam All,the 12th peak is saurucinone,the 13th peak is saurucinol,the 14 peak is likarinA,the 15 peak is 4-O-Demethylmanassantin B and the 17 peak are likarinB,and the 19th peak is manassantin B.The similarity calculation results of 34 batches of Saururus chinensis(Lour.)Baill.fingerprints showed that the similarity of 24 batches of samples was above 0.90,and the similarity of 10 batches of samples was between 0.80 and 0.90.(3)Thin layer chromarography research:Using Saururus chinensis(Lour.)Baill.and flavonoids(rutin,hyperin,Iso-quercitrin,quercitrin)as indicators,a rapid identification method for thin layer chromarography of was established;As a indicator of lignans(likarin B,saurucinone,likarin A),a rapid identification method for thin layer chromarography of Saururus chinensis(Lour.)Baill.was established.(4)Determination of content:The total flavonoids content of 34 bat ches of Saururus chinensis(Lour.)Baill.was between 2.26%and 5.41%;The total ligans content of 34 batches of Saururus chinensis(Lour.)Baill.was between 0.56%and 1.21%;The results showed that the content of 34 batches of rutin in Saururus chinensis(Lour.)Baill.was Between 0.0135%~0.0476%;the hyperoside is between 0.0268%~0.0654%;isoquercitrin is between0.0096%~0.0530%,;quercetin is between 0.0226%and 0.0734%;quercetin is between 0.0019%and 0.0161%;And the total content of five flavonoids ranged from 0.0757%to 0.2642%.The content of saurucinone all of 34 batches of Saururus chinensis(Lour.)Baill.were greater than 0.1%;the content of saurucinol is between 0.0079%and 0.0238%;the Saurucinol I content is 0.0122%~0.0330%;the likarinA content is between 0.0059%~0.1136%;the content of 4-O-Demethylmanassantin B is between 0.0079%-0.0410%;The content of LikarinB is between 0.0010%~0.0371%;The content of ManassantinB is between 0.0151%and 0.0285%;and the total content of six lignans ranged from 0.1000%to 0.2431%except saurucinone.Study on Harvesting Processing Technology and harvesting seasons of Saururus chinensis(Lour.)Baill.(1)Different drying process studies Under different drying conditions,under different drying conditions,the total flavonoid content,total lignan content,total flavonoid content and total lignan content of Saururus chinensis(Lour.)Baill.showed that it was dried at 30℃ in the oven.The retention efficiency is the highest under the conditions.Therefore,the drying process is recommended to choose drying in oven at 30℃.(2)Study on the different lignin components of Saururus chinensis(Lour.)Baill.in different harvesting seasons in different harvesting seasons.On the whole,the total flavonoid content and total flavonoid content of Saururus chinensis(Lour.)Baill.:March>June(flowering period),total lignan content and total lignan content of Saururus chinensis(Lour.)Baill.:June(flowering period)>March;Previous studies have shown that the lignan compound of Saururus chinensis(Lour.)Baill.is an effective part of anti-eb virus,and comprehensive consideration is given to choose June(flowering period)as it.s harvesting season.In conclusionBased on the current edition of the Chinese Pharmacopoeia quality standard,this study conducted a comprehensive quality control study on Saururus chinensis(Lour.)Baill.,and established the HPLC fingerprint of Saururus chinensis(Lour.)Baill.,the rapid identification method of TLC,and the total of Saururus chinensis(Lour.)Baill..The method for detcrmining the content of flavonoids and total lignans,the determination of the uple content of seven lignans and five flavonoids,further improved the quality control level of Saururus chinensis(Lour.)Baill..The investigation showed the
