Objective:To approach the role of polymorphism of MCP-1 genes in the pathogenesis and development of Type 2 diabetes mellitus (T2DM) and its macrovascular and microvaspular complications. Methods:We genotyped 55 normal subjects,64 type 2 diabetes patients without complications,48 type 2 diabetes patients with microvascular complications and 23 type 2 diabetes patients with macrovascular complications.All the subjects were drawed fasting venous blood,then extracted genomic DNA.Genotypes of the MCP-1G(2518) A were performed by polymerase chain reaction followed by digestion with appropriate restriction endonucleases.At the same time,FPG,2hPG,FINS,FC,HbAlC,TC,TG,LDL-C,HDL-C,UA,body heightN body weight were measured and calculated body mass index(BMI).The results of determination were analyzed statistically. The risk factors of T2DM were analyzed by Logistic (Backward Stepwise) Multivariate analysis. Results:(1) MCP-1 gene have three polymorphisms:AA genotype,AG genotype and GG genotype.(2) The frequencies of the GG(-2518) genotype and the frequencies of the MCP-1 G(-2518) allete were significantly higher in patients with T2DM compared with normal controls, and there was statistical significance (P<0.01) .(3) The frequencies of the GG(-2518) genotype were higher in patients with T2DM without diabetic complications and with diabetic microvascular complications compared with normal controls (P<0.05) . The frequencies of the MCP-1 G(-2518) allete were significantly higher in patients with T2DM without diabetic complications and with diabetic microvascular complications compared with normal controls (P< 0.01) , and there was statistical significance. The frequencies of the GG(-2518) genotype and the frequencies of the MCP-1 G(-2518) allete were slightly higher in patients with diabetic macrovascular complications compared with normal controls (34.8%Vs21.8% and 54.3%Vs39.1%) ,but there was no statistical significance (P >0.05) .(4) There were no statistical significance for frequencies of the GG(-2518) genotype and the frequencies of the MCP-1 G(-2518) allete with diabetic complications compared to the uncomplicated (P>0.05) .(5) Multivariate analysis revealed that the G(-2518) allete was the risk factor of T2DM (OR=3.902, 95%CI: 0.99-16.16, P=0.043) . Old age, hypertension, high triglycerides, low high-density lipoprotein, high low-density lipoprotein, high uricacid and HOMA-IR were the risk factors of T2DM, but height,weight,BMI and TC were not the independent risk factors of T2DM.Conclusions:(1) MCP-1 gene have polymorphisms in the Han population of type 2 diabetes in Kunming, China.(2) The frequencies of MCP-1 G (-2518) allele were significantly higher in the Han population of type 2 diabetes in Kunming, China. MCP-1 G(-2518) allete may be the risk factor of T2DM, and polymorphisms of MCP-1 genes may be associated with T2DM.(3) Old age, hypertension, high triglycerides, low high-density lipoprotein, high low-density lipoprotein, high uricacid ,HOMA-IR and high frequencies of MCP-1 G (-2518) allele were the risk factors of T2DM...
