Objective:1.To detect the expression of chloride intracellular channel protein 1(CLIC1)in normal cervical tissues,low-grade cervical squamous intraepithelial lesions(LSIL)tissues,high-grade cervical squamous intraepithelial lesion(HSIL)tissues,and cervical squamous cell carcinoma(CSCC)tissues and compare the differences among them.2.To compare the infection status of high-risk human papillomavirus(HR-HPV)in normal cervical tissues,LSIL tissues,HSIL tissues and CSCC tissues,and analyze the correlation between CLIC1 expression and HR-HPV infection.3.To analyze the correlation between the expression of CLIC1 and the age,tumor size and histological differentiation of cervical cancer patients.Methods:Randomly selected paraffin specimens of normal cervical tissues(20 cases),LSIL tissues(20 cases),HSIL tissues(20 cases)and CSCC tissues(20 cases)from August2016 to December 2018 in the Obstetrics and Gynecology Clinic or ward of the Second Hospital of Shanxi Medical University.The age of theparticipants,HR-HPV infection status,tumor size,clinical stage and degree of histological differentiation of tumor patients were collected.The expression of CLIC1 were detected in all samples by immunohistochemical SP method and the expression levels were compared among four groups.The correlation of CLIC1 expression levels with age,HR-HPV infection status,clinical stage of cervical cancer and degree of tumor differentiation were analyzed.Results:1.The results of immunohistochemistry showed that the positive expression rates of CLIC1 protein in normal cervical tissue,LSIL tissue,HSIL tissue and CSCC tissue increased gradually was 25%(5/20),50%(10/20),65%(13/20)and 80%(16/20).Linear trend test showed that the positive rate of CLIC1 protein expression increased with the increase of cervical lesions(?~2=12.927,P<0.01).2.The Infection rates of HR-HPV in normal cervical tissues,LSIL tissues,HSIL tissues and cervical cancer tissues were 15%(3/20),80%(16/20),90%(18/20)and 85%(17/20).Linear trend test showed that the detection rate of HR-HPV infection increased with the increasing of cervical lesions(?~2=21.787,P<0.01).Spearman correlation analysis showed the expression of CLIC1 was positively correlated with the infection of HR-HPV,and the correlation coefficient was 0.231,P<0.05.3.There was no significant correlation between the expression of CLIC1 protein and tumor size and histological differentiation of the cervical cancer(P>0.05),while the expression of CLIC1 protein was positively correlated with the age of patients,the correlation coefficient was 0.500,P<0.05.Conclusion:1.CLIC1 protein is highly expressed in cervical cancer and precancerous lesions,and is associated with lesion severity and HR-HPV infection.2.The abnormal expression of CLIC1 gene may play a role in the development of cervical cancer.
