Objective:To investigate the effect of karyopherin ?2(KPNA2)on osteosarcoma(OS)proliferation and angiogenesis,and to analyze its possible mechanism at the molecular level.Methods:(1)Saos-2 cells were transfected with Sh-KPNA2 lentiviral vector and empty Sh-Control vector,and the effect of KPNA2 knockout on the proliferation of Saos-2 cells was observed.The growth curve was used to reflect the effect of KPNA2 on Saos-2 cells.(2)OS animal models were constructed using Saos-2 cells transfected with Sh-KPNA2 and the empty Sh-Control vector,and the tumor weight and volume of the two groups of nude mice were compared after tumor formation.(3)The effect of KPNA2 on tumor angiogenesis was verified by fluorescence molecular tomography(FMT)of small animals in vivo before the nude mice were sacrificed.(4)QRT-PCR and western-blot were used to verify the mRNA and protein expression levels of KPNA2 and SRY related HMG box-2(Sox2)genes in the Y chromosome.Results:(1)The proliferation capacity of Sh-KPNA2 group was significantly lower than that of Sh-Control group(P<0.05).(2)The tumor volume and weight of tumor-bearing mice in the Sh-KPNA2 group were smaller than those in the Sh-Control group(P<0.05).FMT scan results showed that the angiogenesis of Sh-Control group was higher than that of Sh-KPNA2 group(P<0.05).(3)Both qRT-PCR and western-blot results of the tumor showed that the expression of Sox2 in Sh-Control was higher than that in Sh-KPNA2.Conclusion:(1)KPNA2 plays a crucial role in the occurrence and development of osteosarcoma.(2)KPNA2 knockout can inhibit OS proliferation and thus inhibiting the development of OS.(3)KPNA2 can promote OS angiogenesis,thus promoting its early metastasis.(4)The expression of KPNA2 in osteosarcoma is closely related to the expression of Sox2.
