Objective:Using modern science and technology,research is carried out on the production specifications and standards of Radix Angelicae Dahuricae pieces.From the aspects of the processing technology of the pieces,the establishment of the quality control system,and the comparative study of the two pieces of decoction,the integration of the traditional Chinese medicine industry in Radix Angelicae Dahuricae was established,and the whole process of the production of the pieces was standardized,and the development of the whole industry of the white peony was promoted.Methods:?1?Orthogonal experimental design combined with multi-index multi-evaluation,star-point experimental design combined with single factor investigation,with active ingredients of Imperatorin,isoimperatorin,oxidized praeruptorin,bergamot lactone,total coumarin The content of total polysaccharide content of Radix Paeoniae Alba is the total index.The water temperature,water addition amount,soaking time,cutting thickness,drying temperature and drying time are taken as the investigation factors.The traditional processing conditions of white peony are preferred,and the water content of fresh medicinal materials,cutting thickness,Drying time and drying temperature are the factors of investigation,and the conditions of white peony fresh cutting are preferred,and the packaging materials of the pieces are screened.?2?According to the"Guiding Principles for Formulating Quality Standards of Traditional Chinese Medicine",Radix Angelicae Dahuricae pieces from different origins were collected for TLC identification,water limit determination,ash limit determination,extract content determination and active ingredient content determination,and the multi-evaluation content determination method and characteristic Atlas of Radix Angelicae Dahuricae were established.?3?The active ingredient content,fingerprint and fat-soluble components were compared between the two pieces;the hot plate method,acetic acid writhing method and xylene method were used to compare the efficacy;the half lethal dose and the maximum tolerated amount were compared for safety comparison.Results:?1?The optimum processing conditions of Angelica dahurica traditional processing technology are 4 times of 25°C water,soaked for 6h,run through,and6mm,60°C,dry for 3 hours.The best process conditions of Angelica dahurica fresh cutting process are water content 35%,cut 6mm,Dry at 60°C for 12 hours.The packing is ordinary plastic atmospheric packing.?2?The quality standard of Angelica dahurica was established.In TLC identification,the oxidized praeruptorin was added as the reference substance.In the chromatogram of the tested substance,the spots of the same color were displayed at the corresponding positions of the chromatogram of the reference substance and the chromatogram of the reference substance.Along the standard of pharmacopoeia,the moisture content of ash and extract should not exceed 14%,the ash content should not exceed 5%,and the extract should be greater than or equal to 15%.The quantity and content of active ingredients should be limited to 0.08%for imperatorin,0.056%for isoimperatorin,0.089%for oxidized imperatorin and 0.013%for bergamot lactone,respectively.Heavy metals are Pb?5.0 mg kg-1,Cd?0.3 mg kg-1,Hg?0.2 mg kg-1,As?2.0 mg kg-1,and Cu?20.0 mg kg-1.Agricultural residue and aflatoxin can not be detected.?3?The content of active ingredients in fresh cut Angelica tablets is higher than that of traditional Angelica tablets.There is no significant difference in fingerprints.The types of fat-soluble components are mostly the same,but there is a certain difference in proportion and content.Both fresh-cut Angelica dahurica decoction tabletsAandAtraditionalAAngelicaAdahuricaAdecoctionatabletsacanaincreaseatheapai nathreshold of mice,reduce theanumber ofawrithing inamice,and reduce the swelling degree of mouse ear slices,so that the effect of fresh-cut Angelica dahurica is better.Interms of safety,the maximum tolerated dose of fresh cut pieces was 177.87g?raw drug?·kg-1?body weight?,and the maximum tolerated amount of traditional pieces was 93.1g?raw drug?·kg-1?body weight?.The former is significantly higher than the latter.Conclusions:This experiment is of great significance to the traditional construction of Angelica dahurica,the fresh-cutting process,the quality standard,and the comparison of the content,efficacy and safety of the two pieces.
