Purpose:To observe the effect of penetrative needling from Baihu(GV20)to Qubin(GB70)(PNBQ)on neurologic functions.The effects of this acupuncture method on ischemic reperfusion injury were investigated based on the regulating mechanism of flammatory cytofactor and endothelial cells adhesion.It provides experimental basis for the study of the pathophysiological mechanism of ischemic cerebrovascular disease and the application of traditional Chinese medicine acupuncture therapy in this field extensively.Methods:Fifty-four SD rats were randomly divided into Sham group,model group,A model of cerebral ischemia reperfusion injury in rats using a monofilament thread method.When rats woke up and their vital signs were stable.Through the neural function score to determine whether the model is successful.Acupuncture group rats were given acupuncture(Baihuito Qubin)on the side of the injured.Acupubcture groups rats received needling therapeutic 30 minutes each time and twice in one day.The neurological function severity was scored respectively 24 h,48 h and72 h for each group after modeling.The changes were compared at the end of the course and the ischemic brain tissue of the rats were observed by microscope and ultrastructure pathology of microvascular endothelial cell were observed by transmission electron microscope.Expression of ICAM-1and VCAM-1 in brain tissue of rats need to be tested by IHC method,also the changes in in serum and brain tissue of rats in respect of IL-6 and TNF-? expression by ELISA method.Results:1.Compared with the model group,the score of nerve function defect in acupuncture group was significantly reduced at 24 h to 72h(P<0.05),;2.Compared with the model group,the number of platform crossings in the acupunctrue group increased significantly(P<0.05)after 72 hours of acupuncture treatment;Acupuncture group escape-period significantly shortened(P<0.05)by 72 h which indicating that acupuncturecan improve the learning and memory ability and upgrate nerve function of rats with cerebral ischemia reperfusion injury;3.Acupuncture can inhibit ICAM-1 and VCAM-1 expression and prevent intercellular adhesion and signal transmission;4.Acupuncture can inhibit the expression of inflammatory factors IL-6 and TNF-? in serum and brain which indicating that acupuncture has an inhibitory function of inflammation;5.Acupuncture can maintain the normal structure of TJ in hippocampal endothelial cells and improve the ultrastructure of nerve cells.Conclusions: 1.Acupuncture can significantly reduce the degree of nerve function defect and ischemic injury of hippocampus neurons in rats and the learning and memory ability and nerve function in rats with cerebral ischemia reperfusion injury were promoted.2.Acupuncture can inhibit nerve inflammation which is related to inhibiting the expression of inflammatory factorsIL-6?TNF-? and reducing the expression of ICAM-1?VCAM-1 protein,preventing intercellular adhesion and signal transmission.
