Objective:To study the expression of Prospero-related homeobox 1(Prox-1)and Cell adhesion molecule variant 6(CD44v6)in gallbladder carcinoma(GBC),adenomatous polyps of gallbladder and chronic cholecystitis,and to analyze the relationship between the expression of Prox-1 and CD44v6 proteins in gallbladder carcinoma and their clinicopathological features,as well as the correlation between the positive expression of Prox-1 and CD44v6 in gallbladder carcinoma,and the clinical significance of the two factors in gallbladder carcinoma.Methods:Fifty-eight paraffin tissue specimens of primary gallbladder cancer diagnosed by WHO pathological diagnostic criteria were collected from the Department of Hepatobiliary Pancreas and Spleen Surgery,Affiliated Hospital of Inner Mongolia Medical University from July 2010 to April 2018.In addition,40cases of adenomatous polyps of gallbladder and 60 cases of chronic cholecystitis were selected as controls.The expression of Prox-1 and CD44v6 in the above three tissues were detected by S-P immunohistochemical method,and the differences were compared.The relationship between the expression levels of Prox-1 and CD44v6 in GBC tissues and clinicopathological features was analyzed.The correlation between the expression of Prox-1 and CD44v6 in gallbladder carcinoma tissues was also analyzed.Results:The positive rates of Prox-1 and CD44v6 in gallbladder carcinoma tissues were 62.1%(36/58)and 67.2%(39/58),respectively,which were significantly higher than those in adenomatous polyps of gallbladder and chronic cholecystitis(P<0.05).There was no significant difference in the positive rates of Prox-1 and CD44v6 in gallbladder carcinoma patients with different gender,age and location of cancer occurrence among the clinicopathological features(P>0.05).The positive rates of Prox-1 and CD44v6 in gallbladder carcinoma patients with high,medium and low differentiation were significantly different(P<0.05),and the positive rates in low differentiation group were higher than those in medium and high differentiation group.The lower the differentiation degree,the higher the malignancy degree,the higher the expression intensity in gallbladder carcinoma tissue specimens,and the more positive the two factors in medium and low differentiation group were.The positive rates of Prox-1 and CD44v6 in gallbladder carcinoma patients with different TNM stages were significantly different(P<0.05),and the positive rates of stage III-IV were higher than those of stage I-II.The positive rates of Prox-1 and CD44v6 in gallbladder carcinoma patients with or without lymph node metastasis were significantly higher than those in non-metastasis group(P<0.05).Spearman correlation test showed that Prox-1 was positively correlated with CD44v6 in gallbladder carcinoma(r=0.363,P=0.005).Conclusion:Prox-1 and CD44v6 are highly expressed in gallbladder carcinoma tissues,which are closely related to lymph node metastasis,differentiation and TNM staging.Prox-1 and CD44v6 are positively correlated in gallbladder carcinoma,and are associated with the occurrence and development of gallbladder carcinoma and abnormal expression of both.The combined detection of Prox-1 and CD44v6 has certain clinical significance.It has important clinical significance in evaluating and judging gallbladder carcinoma lymph node metastasis and malignancy.
