| Objective Research from the microscopic point of view through experiments to evaluate the therapeutic effect of Jia Wei Huang Qi Jian Zhong decoction on chronic atrophic gastritis(CAG)by Professor Zhu Zongyuan,a famous veteran Chinese medicine in Inner Mongolia Autonomous Region.Methods SD rats of clean grade were selected,half male and half female.Random number table method: normal group,model group,control group,high,medium and low dose group of Jia Wei Huang Qi Jian Zhong decoction,8 in each group,a total of 48.CAG animal model was established by stimulating gastric mucosa with free drinking MNNG in rats;the treatment group was treated with high,medium and low doses of modified Jia Wei Huang Qi Jian Zhong decoction;the control group was treated with Vitamycin suspension;the normal group and model group were not treated with normal diet.After the experiment,the histopathological changes of gastric mucosa were observed by HE staining,and the expressions of PCNA(BCL2-Associated X),bax(BCL2-Associated X)and bcl-2(B-cell lymphoma-2)in gastric mucosa were detected by immunohistochemistry.Result 1.Histopathological observation of gastric mucosa under light microscopy:Compared with the normal group,there were pathological changes of thinning of gastric mucosa,disappearance of mucosal epithelium and intrinsic glands,thickening of muscular mucosa and infiltration of a large number of inflammatory cells in each group after modeling.Compared with the model group,each treatment group could improve the intrinsic layer and mucosal epithelium of gastric mucosa and reduce the infiltration of inflammatory cells.The middle and high dose groups of Jia Wei Huang Qi Jian Zhong decoction had more obvious effects on the arrangement of mucosal glands and the thickening of mucosa.2.Weight analysis of rats in each group: Compared with the normal group,the weight of each group decreased(P<0.05),the weight of the treatment groupincreased(P<0.05)compared with the model group,and the weight of the middle and high dose groups of Jia Wei Huang Qi Jian Zhong decoction increased significantly(P<0.05)compared with the model group,with statistical significance.3.The expression of PCNA,bax and bcl-2 in gastric mucosa of rats in each group: Compared with the normal group,the expression of PCNA and bcl-2 in model group and control group increased,while the expression of bax decreased,with statistical significance(P<0.05).Compared with the model group,the low,medium and high dose groups of the control group and the modified Jia Wei Huang Qi Jian Zhong decoction could reduce the expression of PCNA,with statistical significance(P<0.05);compared with the control group,the middle and high dose groups of the modified Jia Wei Huang Qi Jian Zhong decoction played a significant role in the down-regulation of PCNA expression,with statistical significance(P<0.05).Compared with the model group,the high,medium and low dose groups of Jia Wei Huang Qi Jian Zhong decoction could significantly up-regulate the expression of bax(P<0.05);compared with the control group,the middle and high dose groups of Jia Wei Huang Qi Jian Zhong decoction had significant up-regulation effect on bax(P<0.05).Compared with the model group,the low,medium and high dose groups of the control group and the modified Jia Wei Huang Qi Jian Zhong decoction all had a significant decrease in bcl-2expression(P<0.05);compared with the control group,the medium and high dose groups of the modified Jia Wei Huang Qi Jian Zhong decoction played a significant role in the down-regulation of bcl-2 expression(P<0.05).Conclusion Jiawei Huangqi Jianzhong decoction has a good therapeutic effect on chronic atrophic gastritis.The microcosmic mechanism of Jiawei Huangqi Jianzhong Decoction may be related to regulating the expression of PCNA,Bax and bcl-2,improving the histomorphological changes of gastric mucosa,regulating the proliferation of gastric mucosal cells and the imbalance of apoptosis.The specific pathway of action needs further study. |
