Objective:Oenanthe javanica is a herbaceous plant of the genus Umbelliferae,which is a special plant of traditional Chinese medicine and food.As a medicine and food plant,as early as in ancient times,people have been used as top grades in vegetables.At present,there are few reports on the efficacy and mechanism of sedative and hypnotic in Oenanthe javanica.This experiment first studied the optimal extraction process of total Oenanthe javanica extract.The animal experiment was used to screen the effective part of the sedative and hypnotic effect of the Oenanthe javanica and to preliminary explore its mechanism.At the same time,the chemical composition of the effective part was initially analyzed.Finally,the mechanism of intervention of insomnia in the effective part of Oenanthe javanica is clarified.It provides a theoretical basis and scientific basis for the development of new functional foods or drugs for Oenanthe javanica,which can expand the source of tranquillizing Chinese medicine and enable more comprehensive development of Oenanthe javanica resources.Methods:The response surface method was used to optimize the extraction process of total Oenanthe javanica extract.The sedative hypnosis model induced by pentobarbital sodium and a PCPA-induced insomnia model were established to screen the effective part of sedative hypnosis of Oenanthe javanica and to preliminary explore the mechanism of sedative and hypnotic effects of Oenanthe javanica extract.The chemical composition of the effective part of Oenanthe javanica was analyzed by UPLC-Q-TOF-MSE technology combined with UNIFI platform.The non-targeted metabolomics technique was used to analyze the effect of the effective part of Oenanthe javanica on the small molecule metabolites in the body,and the metabolic pathway of Oenanthe javanica intervention in insomnia was studied.Results:According to the mathematical model established by response surface analysis,the optimal extraction conditions were found to be:extracting for ethanol concentration of 71.91%and liquid-to-solid ratio of 10.91ml/g for 3.5 h at 71.18?.Validation Under this condition,the predicted paste yield is 17.79%.In each extract of Oenanthe javanica,ethyl acetate extract can significantly increase the time of sleep and sleep rate of mice.Ethyl acetate extract of Oenanthe javanica can improve the general physiological condition of insomnia rats,increase the content of 5-HT,TNF-?and IL-1?,and increase the expression of 5-HT1aa receptor mRNA.A preliminary analysis of the ethyl acetate extract from the water was found to identify 11 compounds.Twenty-three significant differential metabolites after insomnia were detected by metabolomics techniques.After the intervention of the effective part of the Oenanthe javanica,seven metabolites were metabolized.These metabolites mainly affect the body's glycerophospholipid metabolism,tryptophan metabolism,pantothenic acid and CoA biosynthesis,?-alanine metabolism,lysine biosynthesis,phenylalanine metabolism,pyrimidine metabolism,arginine and guanidine metabolic pathways.Conclusion:The ethyl acetate extract of Oenanthe javanica is an effective part of sedative and hypnotic effect of Oenanthe javanica,which can increase the content of 5-HT,TNF-?and IL-1?,and increase the expression of 5-HT1aa receptor mRNA.It mainly intervene the insomnia by affecting the body's metabolic pathways such as glycerophospholipid metabolism,tryptophan metabolism,pantothenic acid and CoA biosynthesis,?-alanine metabolism.
