Objective: This study aimed to detect the expression of transferrin receptor 1(TfR1)and vascular endothelial growth factor(VEGF)in osteosarcoma,and analyze its relationship with clinicopathological features and prognosis of osteosarcoma patients,the correlation of its expression in osteosarcoma tissue and explore its clinical significance.Methods: The current study included 35 patients who underwent surgical resection and pathological diagnosis was osteosarcoma between January 2011 and December 2015 in The Fourth Hospital of Hebei Medical University.The lesions and para-carcinoma tissues were collected.The expression of TfR1 and VEGF in specimens was detected by immunohistochemical staining.The relationship between the expression levels of TfR1 and VEGF and clinical features was analyzed by Chi square test or Fisher exact probability method.Kaplan-Meier curve was used for survival analysis.Spearman correlation analysis used to explores the correlation between TfR1 and VEGF.P<0.05 was considered statistically significant.Results: 1.Expression of TfR1 and VEGF in osteosarcoma and paracancerous tissues: TfR1 expression is mainly localized in the cell membrane and cytoplasm near the cell membrane,the moderate-high expression rates in osteosarcoma and para-carcinoma tissues were 74.3%(26/35)and 20%(7/35),respectively,and the difference was statistically significant(P<0.05);VEGF expression is primarily localized in the cytoplasm of cells,the moderate-high expression rates in osteosarcoma and para-carcinoma tissues were 77.2%(27/35)and 11.4%(4/35),respectively,and the difference was statistically significant(P<0.05).2.The relationship between TfR1 and VEGF expression in osteosarcoma and clinicopathological features: The expression of TfR1 was associated with histological differentiation,distant metastasis and Enneking staging(P<0.05),but not with gender and tumor size(P>0.05).The expression of VEGF was correlated with histological differentiation,distant metastasis and Enneking staging(P<0.05),but not with gender and tumor size(P>0.05).3.The results of Kaplan-Meier survival analysis showed that patients with low expression of TfR1 and VEGF had longer survival than that of moderate-high expression.The difference was statistically significant(P<0.05).4.Spearman correlation analysis showed that there was a significant positive correlation between TfR1 and VEGF expression in osteosarcoma(r=0.669,P<0.05).Conclusions: 1.TfR1 and VEGF play an important role in the development,invasion and distant metastasis of osteosarcoma,which may indicate the degree of malignancy of the tumor,and it is related to the prognosis of osteosarcoma.2.TfR1 and VEGF may play a synergistic role in the development of osteosarcoma.3.TfR1 can be used as an auxiliary diagnosis for osteosarcoma,to determine the degree of malignancy,whether distant metastasis occurs,to evaluate prognosis,and to provide new targets for its treatment.
