Objective:To observe the microRNA(miRNA,miR)-424-3p and 5p in prostate cancer cell line LNCaP expression and its impact on the proliferation and migration and then contrast miR-424-3p and 5p on biological behavior influence of LNCaP whether there are different.Methods: Nuclear small RNA U6(snRNA U6)as a internal reference,real-time fluorescence quantitative polymerase chain reaction(FQ-PCR)was used to detect relative expression of miR-424-3p and 5p in the RWPE-1 and LNCaP cells.miR-424-3p and 5p inhibitor(antisense RNA)respectively were transfected into LNCaP cells through Application of transient transfection technology.Cell counting Kit(CCK-8),flat plate cloning test were used to detected cells proliferation.Transwell assay was performed to detect the cell invasion ability change and flow cytometry tested cell apoptosis and cell cycle changes,ultimately compare the effect of miR-424-3p and 5p on LNCaP cells biological behavior.Results: Real-time fluorescence quantitative polymerase chain reaction showed that the expression of miR-424-3p and 5p in LNCaP cells was higher than that in RWPE-1 cells(Human normal prostate epithelial cells),the difference was statistically significant(P<0.05);miR-424-3p and 5p expression in RWPE-1 cells was no statistically significant difference(P>0.05),but the relative expression of miR-424-5p was higher than 3P in LNCaP cells,the difference was statistically significant(P<0.05);Cell counting Kit(CCK-8),colony formation test and Transwell test showed the proliferation and migration was significant inhibited,the difference was statistically significant(P<0.05);Flow cytometry showed increased proportion of cells in early apoptotic cells,increased the proportion of cells in S phase,the difference was statistically significant(P <0.05).The effects of miR-424-3p and 5p on the biological behavior of LNCaP cells were compared,and the differences were not statistically significant(P>0.05).Conclusion:1.The expression of miR-424-3p and 5p in prostate cancer cell line LNCaP is higher than RWPE-1 cells.2.After MiR-424-3p and 5p inhibitors(Antisense oligonucleotide)were transfected respectively,the proliferation and migration of LNCaP cells obviously were inhibited and the proportion of early apoptotic cells increased,but both on LNCaP prostate cancer cell biological behavior were no obvious difference.So we hypothesized that the influence of miR-3p and 5p on tumor cell biological behavior are not necessarily proportional with their expression abundance.Because both has an significant impact on the biological behavior of proliferation and migration of prostate carcinoma LNCaP cells,we can further speculate that the effect of suppression of miR-3p and 5p together or the common pre-RNA may be better.
